A alcachofra (Cynara scolymus L.) é uma espécie vegetal conhecida desde a antiguidade, preconizada por suas ações coleréticas e diuréticas. A ação farmacológica e terapêutica das folhas de alcachofra não é devida exclusivamente à cinarina, mas a um conjunto de constituintes quimicamente semelhantes denominados polifenóis. Muitos métodos analíticos são citados na literatura para a quantificação do teor de cinarina ou de ácido clorogênico presente na folhas de alcachofra ou em preparações farmacêuticas. Neste trabalho foram utilizadas duas metodologias oficiais: da Farmacopéia Européia 5. edição (2006) e a da Farmacopéia Brasileira 3. edição (1977). Uma terceira metodologia foi desenvolvida a validada, utilizando o reativo de Arnow como agente complexante dos polifenóis totais. Uma amostra de extrato seco de alcachofra e três amostras de fitoterápicos encontradas no mercado foram analisadas. Na amostra de extrato seco analisada pela metodologia da Farmacopéia Européia foi encontrado um teor de ácido clorogênico (0,16 por cento) bem inferior ao mínimo estabelecido pela própria Farmacopéia de 0,8 por cento para as folhas de alcachofra. O método espectrofotométrico da Farmacopéia Brasileira mostrou-se inexato e com grande variação de resultados encontrados. O método com o reativo de Arnow foi considerado linear, exato e repetitivo e de menor custo, rapidez e praticidade quando comparado com o método por CLAE da Farmacopéia Européia.
Abstract
Artichoke (Cynara scolymus L.) is a vegetal species known since antiquity, praised for its choleric and diuretic actions. The pharmacologic and therapeutic actions of the artichoke leaves aren’t exclusively related to cinarine, but also to a group of constituents with similar chemistry called polyphenols. Some analytical methods are cited in literature to quantify the cinarine or chlorogenic acid content in the leaves of the artichoke or in pharmaceutical preparations. In this work, two official methodologies were used, one from European Pharmacopeia 5th edition (2006) and another from Brazilian Pharmacopeia 3rd edition (1977). A third methodology was developed and validated, using the Arnow reactive as complexant agent of the total polyphenols. A sample of dry extract of artichoke and three samples of phytotherapics found in the market were analyzed. The spectrophotometric method of the Brazilian Pharmacopeia was inexact and showed great variation of results. The proposed method was considered linear, accurate, reproducible and cheaper, faster and more practical when compared with the European Pharmacopeia method.
