EFEITO DA TEMPERATURA NA INFECÇÃO DE RHODNIUS PROLIXUS POR TRYPANOSOMA CRUZI E TRYPANOSOMA RANGELI

A temperatura ambiental afeta diversos processos bioquímicos, fisiológicos e comportamentais, especialmente em animais ectotérmicos. Mudanças nesse parâmetro podem alterar o fitness de insetos vetores e sua distribuição no ambiente, modificando consequentemente, a dinâmica de transmissão de doenças. O objetivo do presente estudo foi avaliar os efeitos da temperatura no desenvolvimento da infecção de Trypanosoma cruzi e Trypanosoma rangeli em Rhodnius prolixus. Os ensaios com T. rangeli foram desenvolvidos com parasitos submetidos a dois tratamentos.
T. rangeli T1 foram obtidos através de passagens em LIT por três meses. T. rangeli T2 foram mantidos em LIT por um mês após uma passagem triatomíneo-camundongo. Inicialmente, o crescimento dos parasitos em meio de cultura submetidos a diferentes temperarturas (21, 24, 27 e 30°C) foi avaliado. O crescimento do T. cruzi foi termodependente, com um aumento de 28 vezes da população mantida em 30°C ao final de sete dias. A mortali dade ficou abaixo dos 10%, exceto para os parasitos mantidos a 21°C, onde esse valor ficou próximo aos 20%. Para os dois tratamentos de T. rangeli , o crescimento máximo foi observado a 27°C. Entretanto, as populações de T. rangeli T2 cresceram mais do que as de T. rangeli T1 e apresentaram taxas de mortalidade mais baixas, em torno de 5%. O próximo objetivo foi avaliar diferentes parâmetros fisiológicos de R. prolixus infectados pelos parasitos e submetidos às mesmas condições de temperaturas. A infecção por T. cruzi prolongou o período intermudas em todas as temperaturas e reduziu o número de insetos aparentemente saudáveis em 20%. A virulência do parasito foi dependente da temperatura e do estado nutricional dos insetos. T. rangeli T1 foi mais virulento do que T. rangeli T2, particularmente nas temperaturas mais baixas. Finalmente, o crescimento do T. cruzi no inseto foi quantificado por qPCR e câmara de Neubauer. Os resultados, ainda preliminares, corroboraram com os dados do crescimento do parasito em LI T. De maneira geral, os resultados sugerem que os dois parasitos podem ser patogênicos ao vetor na natureza dependo das condições a que são submetidos.
