ANÁLISES MULTILOCUS DE LEISHMANIA INFANTUM (KINETOPLASTIDAE: TRYPANOSOMATIDAE) E DO COMPLEXO LUTZOMYIA LONGIPALPIS (DIPTERA: PSYCHODIDAE) NO BRASIL

Lutzomyia longipalpis (Lutz & Neiva, 1912) é a principal espécie transmissora da Leishmaniose Visceral Americana (LVA) e apresenta ampla distribuição geográfica que se estende do México ao Norte da Argentina. Esse vetor constitui na verdade um complexo de espécies crípticas fortemente suportado por estudos comportamentais, bioquímicos e moleculares. O gene period (per) tem se mostrado um bom marcador molecular na detecção dessas espécies e, junto à outros marcadores como som de cópula e feromônios, consolidou a existência do complexo L. longipalpis no Brasil. Contudo não se sabe quais são e como se distribuem essas espécies, e nem se elas apresentam relação diferenciada com Leishmania chagasi, agente etiológico da LVA, ou com a dinâmica de transmissão dessa doença. As populações de L. longipalpis de duas microrregiões do município de Pancas (ES), delimitadas pela topografia da região, foram comparadas molecularmente a partir de seqüências do gene per e apresentaram baixa estruturação com fluxo gênico restrito entre elas. Por outro lado, as evidências de introgressão entre espécies simpátricas em Sobral (CE) e as características desse processo em outros modelos de especiação, como em Drosophila e no complexo A. gambiae, mostram a necessidade da realização de análises multilocus para melhor entender as forças evolutivas envolvidas nesse processo de especiação incipiente. Foram então seqüenciados 12 genes nucleares das populações alopátricas de L. longipalpis da Lapinha (MG) e de Pancas, nas quais o tipo de feromônio, o som de cópula e a divergência genética no gene per mostram representarem duas espécies diferentes do complexo. As análises dos 12 genes suportam a existência de espécies crípticas, contudo, o grau de estruturação mostrado por cada um dos 12 genes, além do gene per, foi bastante variado (0,17 < FST < 0,95). Além disso, a presença de polimorfismos compartilhados entre as populações reforça a idéia de um processo de especiação recente. É importante que sejam realizadas novas análises como, por exemplo a construção de redes de haplótipos ou utilização do modelo de “isolamento com migração” implementado no programa IM, para tentar entender melhor quais as forças evolutivas que atuam no processo de especiação incipiente de L. longipalpis, além da inclusão de outras populações nas análises.
