Os peptídeos antimicrobianos têm se mostrado uma boa alternativa para as terapias antiparasitárias. Neste trabalho descrevemos a estrutura e a atividade biológica da temporizina (FLPLWLWLWLWLWKLK) e sua ação contra formas epimastigotas do Trypanosoma cruzi. A temporizina é um peptídeo híbrido, sintetizado a partir de outros três peptídeos antimicrobianos: a temporina A (FLPLIGRVLSGIL-NH2), a sapecina B (KLKLLLLLKLK-NH2) e a gramicidina A (HCO-VGALAVVVWLWLWLW-NHCH2). Os efeitos da temporizina sobre o Trypanosoma cruzi e sua toxicidade para células de mamíferos foram analisados utilizando as metodologias de citometria de fluxo, microscopia de fluorescência e liberação da lactato desidrogenase (LDH). O valor concentração efetiva de temporizina para lisar 50% das formas epimastigotas de Trypanosoma cruzi (EC50), foi determinado em um ensaio de viabilidade celular através do método de metabolização do brometo azul de tetrazólio (MTT). Para determinar a liberação de hemoglobina e quantificar a porcentagem de hemólise causada pela temporizina em doses letais para o Trypanosoma cruz. Utilizamos o método colorimétrico. Os dados sobre a EC50 e a capacidade hemolítica da temporizina foram então utilizados para a determinação do índice terapêutico do peptídeo. O mapeamento da estrutura da temporizina e a determinação de suas características foram obtidos através da metodologia de dicroísmo circular em um ambiente hidrofóbico que mimetizou as bicamadas das biomembranas. Submetemos a sequência da temporizina à plataforma Itasser para modelagem de proteínas e softwares, com o objetivo de realizar predições teóricas sobre a sua estrutura e características conformacionais. Para avaliar os efeitos da temporizina sobre a eletrofisiologia de membranas, foi usada a técnica de Patch-Clamp em célula inteira ou célula ligada. Essa técnica associada com experimentos envolvendo captação de corantes com diferentes valores de massa molecular permitiu determinar o limite de exclusão dos poros formados pelo peptídeo. Tendo em vista, as características estruturais e a atividade tripanocida da temporizina, concluímos que este peptídeo é uma alternativa viável para o desenho racional de um novo fármaco para o controle da doença de Chagas.
Abstract
The antimicrobial peptides have been shown to a good alternative for the therapy antiparasites. At this work, we describe the structure and the biological activity of temporizin (FLPLWLWLWLWLWKLK) and its action against epimastigotes of Trypanosoma cruzi. The temporizin is a hybrid peptide, synthesized from three other antimicrobial peptides: the temporin A (FLPLIGRVLSGIL-NH2), the sapecin B (KLKLLLLLKLK-NH2) and gramicidin A (HCO-VGALAVVVWLWLWLW-NHCH2). The effects of temporizin over T. cruzi and its toxicity for the mammalian cells were analyzed using flow cytometry, fluorescence microscopy and release of lactate dehydrogenase (LDH). The value of the effective concentration of temporizin to lyse 50% of epimastigotes forms of T. cruzi (EC50) was determined in a cell viability assay using the method of metabolism of tetrazolium blue thiazol methyl bromide (MTT). To determine the release of hemoglobin and to quantify the percentage hemolysis caused by lethal doses temporizin T. cruzi, we used the method colorimetric. The data about the EC50 and the hemolytic capacity of temporizin were then used to determine the peptide’s therapeutic index. The mapping of the structure of temporizin and the determining the characteristics were obtained through the method of circular dicroism (CD) in a hydrophobic environment that mimicked the bilayers of membranes. We submitted the sequence of the platform itasser and software for modeling proteins, with the purpose to make of theoretical predictions about the structure and conformational characteristics. To evaluate the effect of temporizin over membrane electrophysiology, was used the technique of patch-clamp in a whole cell or attached cell form. This technique associated with experiments involving uptake of dyes with different molecular mass values, allowed to determine the exclusion limit of the pores formed by the peptide. Given the structural characteristics and trypanocidal activity of temporizin, we concluded that this peptide is a viable alternative to the rational design of a new drug for the control of Chagas disease.
