Os Staphylococcus são bactérias do grupo dos cocos Gram positivos que fazem parte da microbiota, mas que podem provocar doenças. O aumento das condições que induzem à internação de indivíduos cada vez mais graves e imunocomprometidos, somado ao surgimento da resistência a antimicrobianos, faz com que o uso de antimicrobianos de última linha seja cada vez mais recorrente. O objetivo desse estudo foi identificar os mecanismos de resistência antimicrobiana em bactérias do gênero Staphylococcus associados à infecções clínicas em pacientes assistido em hospital terciário do Recife/PE. A identificação dos isolados foi feita por MALDI TOF. O perfil de sensibilidade aos antimicrobianos foi determinado pela técnica de microdiluição em caldo. Os principais genes de resistência aos antimicrobianos foram identificados por PCR e pelo sequenciamento de DNA. Os determinantes genéticos pesquisados incluíram os cassetes SCCmec tipo I, II, III, IV, V e os genes mecA, que conferem resistência à oxacilina, os genes vanA, vanB, que conferem resistência à vancomicina e os genes cfr e mprF, responsáveis por conferir resistência à linezolida e daptomicina, respectivamente. Foram coletados 36 isolados de Staphylococus aureus e 34 de Staphylococcus coagulase negativa (CoNS), resultando em um total de 70 amostras bacterianas. A resistência à oxacilina em S. aureus foi de 75%, com predominância dos cassetes SCCmec tipo II (37%). Entre os CoNS, o índice de isolados resistentes foi de 85,2%, com predominância dos cassetes SCCmec IV (29%). A frequência do gene mecA nos isolados avaliados foi de 88,2% das amostras. Não foram detectados elementos genéticos de resistência à vancomicina, linezolida e daptomicina. Entretanto, 4,2% das amostras apresentaram baixa susceptibilidade à vancomicina e linezolida. A presença desses isolados reforça a necessidade do desenvolvimento de novas estratégias para combater a ameaça da resistência antimicrobiana aos antibióticos de última linha.
