A letalidade da raiva está próxima de 100% e quinze milhões de pessoas recebem profilaxia pós-exposição anualmente. É imperioso garantir a qualidade dos produtos biológicos utilizados na prevenção desta zoonose. Os lotes de vacina têm a potência avaliada pelo teste do National Institutes of Health (NIH), baseado em imunização e desafio de camundongos. Este ensaio possui várias desvantagens, como a variabilidade significativa e problemas de bem-estar animal. Métodos in vitro, como o ensaio imunoenzimático (ELISA), têm sido propostos com base na qualidade e quantidade da glicoproteína (GLPTN) do vírus da raiva, mas também podem apresentar limitações como baixa sensibilidade, instabilidade e imprecisão. A estimativa da imunogenicidade baseada no título de anticorpos neutralizantes (AN), avaliada por um teste sorológico, como o Teste de Inibição de Foco Fluorescente Rápido Modificado (RFFITm), ainda não é aplicada de forma efetiva para a vacina humana. No entanto, uma concentração de AN pode ser usada como um preditor da eficácia clínica deste produto em humanos vacinados, o que pode ser aplicado na estimativa da potência da vacina. O objetivo deste estudo foi desenvolver e pré-validar um Ensaio Sorológico (ES) utilizando o RFFITm para determinar a potência de vacinas antirrábicas para uso humano, demonstrando sua Relevância e Confiabilidade. Verificar o limite inferior de imunogenicidade ao conteúdo viral de GLPTN capaz de induzir resposta em camundongos, utilizando RFFITm. O ensaio foi capaz de distinguir entre lotes potentes e subpotentes de vacinas. Houve concordância entre as potências determinadas pelo ES e o teste NIH. Os resultados demonstraram que o ES é um candidato confiável para a validação e inclusão em farmacopeias, obtendo redução e refinamento para o teste NIH.
