AVALIAÇÃO DA ATIVAÇÃO DE CÉLULAS ENDOTELIAIS NA INFECÇÃO POR TOXOPLASMA GONDII E A PARTICIPAÇÃO DE VESÍCULAS EXTRACELULARES NO CONTEXTO INFECCIOSO

Toxoplasma gondii é um protozoário pertencente ao filo Apicomplexa identificado pela primeira vez em 1908. Ele é capaz de invadir praticamente todas as células nucleadas, dentre elas encontram-se as células endoteliais. Células endoteliais, além de constituir a principal barreira entre os elementos do sangue e os tecidos, também possuem forte vínculo com o sistema imune. Elas respondem a diferentes estímulos como alterações vasculares, agentes infecciosos ou vesículas extracelulares (VEs) secretadas por diferentes células ou mesmo agentes invasores. Objetivo: avaliar a ativação e disfunção de células endoteliais humanas no contexto da infecção por T. gondii. Material e Métodos: foram cultivadas células HBMEC (células endoteliais da microvasculatura cerebral humana) e HUVEC (células endoteliais do cordão umbilical humano) as quais foram infectadas por T. gondii. A partir disso, foi avaliada a expressão de genes relacionados à ativação/disfunção endotelial e, a nível proteico, a expressão de ICAM-1 e VCAM-1. VEs isoladas de células endoteliais infectadas com T. gondii foram caracterizadas e a carga parasitária de células endoteliais estimuladas com essas VEs foi analisada. Por fim, foi realizada a análise proteômica de VEs produzidas por células endoteliais e por T. gondii. Resultados: células endoteliais apresentaram taxas de infecção e carga parasitária distintas. Na avaliação transcricional, foi observado um aumento de expressão de IL6, IL8, SOCS3, MCP1, VCAM1 e VEGF apenas nas células HUVEC. Células HUVEC infectadas expressaram ICAM-1 e VCAM-1 ao passo que células HBMEC expressaram apenas ICAM-1. Foi observado um aumento na infecção de 14,95% para 22,22% em células HBMEC estimuladas com VEs de T. gondii. Quando as mesmas células foram estimuladas com VEs de HBMEC infectadas a taxa de infecção foi reduzida para 5,36%. Na análise proteômica, foi observado que VEs de células HBMEC infectadas apresentaram proteínas relacionada às junções de ancoragem, aderente e célula-substrato. Discussão: Enquanto células HUVEC apresentaram um perfil pró-inflamatório, células HBMEC apresentaram uma resposta menos intensa. Além disso, mesmo com uma resposta à infecção mais discreta, células HBMEC apresentaram uma menor taxa de infecção do que células HUVEC e ao serem estimuladas com VEs de células infectadas, houve uma redução na taxa de infecção. Conclusão: células endoteliais de diferentes linhagens respondem de forma distinta à infecção por T. gondii.
