Dentre fatores importantes para a evolução da Leishmaniose Tegumentar Americana (LTA)
está espécie do parasito, sendo a caracterização da mesma, de suma importância. Derivados
do RNA ribossomal (rRNA) foram utilizados em estudos para caracterização de Leishmania
spp. através de PCR quantitativa em Tempo Real (qPCR), porém, a região Intergenic Spacer
(IGS) não foi explorada para este fim. Assim, foi proposto a análise in silico e verificação da
aplicabilidade da região IGS rRNA de Leishmania spp. para identificação de espécies que
causam a LTA. Com base em sequências de Leishmania major, foram desenhados primers
para amplificação da região IGS rRNA completa de Leishmania spp. Os produtos foram
submetidos ao Sequenciamento Capilar e de Nova Geração. As sequências obtidas foram
alinhadas e analisadas em relação à tamanho e similaridade, bem como depositadas no
GenBank. Foram analisadas características do elemento de repetição (IGSRE) presente no
IGS rRNA. Além disso, foi desenvolvido e otimizado um sistema primers para identificação
de Leishmania braziliensis para qPCR, sendo aplicado testes de sensibilidade (S),
especificidade () e eficiência (). Verificou-se que o tamanho médio para a região IGS rRNA
é de 3 kb, e a região repetitiva IGSRE varia entre 61-71 pb. A análise de similaridade das
sequências obtidas demonstrou alta conservação entre as espécies. Foram depositadas no
GenBank, quinze sequências parciais geradas para da região IGS rRNA de nove espécies. O
sistema de primers específico para L. braziliensis, apresentou S= 10fg, = 99,73% e, Log =
103
para Leishmania naiffi, Log = 104
para Leishmania guyanensis e Log = 105
para
Leishmania shawi. O sistema pode ser utilizado para diagnóstico, identificação e
quantificação, auxiliando no direcionamento para uma conduta terapêutica mais apropriada
aos casos de infecção por este parasito. E, as sequências inéditas depositadas em bancos de
dados, podem ser utilizadas para múltiplas análises de pesquisadores no mundo.
Resumen en ingles
Among important factors for the evolution of American Cutaneous Leishmaniasis (ACL) is a
parasite species, and its characterization is of paramount importance. Derivatives of ribosomal
RNA (RNAr) were used in studies to characterize Leishmania spp. using quantitative Real
Time PCR (qPCR), however, the Intergenic Spacer region (IGS) was not explored for this
purpose. Thus, the objective of the project was the in silico analysis and verification of the
applicability of the Leishmania spp. IGS rRNA region to identify species that cause LTA.
Based on Leishmania major sequences, primers were designed for complete IGS rRNA
region of Leishmania spp amplification. The products were submitted to Capillary and New
Generation Sequencing. The sequences obtained were aligned and analyzed for size and
similarity, as well as deposited in GenBank. Characteristics of the repeat element (IGSRE)
present in the IGS rRNA were analyzed. In addition, a primers system for identification of
Leishmania braziliensis for qPCR was developed and optimized. Sensitivity (S), specificity
() and efficiency () tests were applied. It has been found that the mean size for the IGS
rRNA region is 3 kb, and the IGSRE repeating region ranges from 61-71 bp. The similarity
analysis of the sequences obtained showed high conservation among the species. Fifteen
partial sequences generated for the IGS rRNA region of nine species were deposited in
GenBank. The specific primer system for L. braziliensis showed S = 10fg, = 99.73% and,
Log = 103
for Leishmania naiffi, Log = 104
for Leishmania guyanensis and Log = 105
for
Leishmania shawi. The system can be used for diagnosis, identification and quantification,
assisting in the targeting of a therapeutic course more appropriate to the cases of infection by
this parasite. And, the unpublished sequences deposited in databases can be used for multiple
analyzes of researchers in the world.
