A garantia do desempenho analítico pode ser verificada através da participação do laboratório em ensaios de proficiência (EP), que são considerados ferramentas externas de garantia da qualidade. Os EP consistem no uso de comparações interlaboratoriais, com o propósito de avaliar a habilidade de laboratórios em realizar um determinado ensaio ou medição de modo competente e demonstrar a confiabilidade dos resultados gerados. O presente trabalho teve como objetivo produzir lotes de itens de ensaio (IE) para serem utilizados em duas rodadas de EP nos ensaios de contagem de bactérias mesófilas e pesquisa de Escherichia coli em água de abastecimento. Os procedimentos de preparo dos IE e as análises de controle foram realizados no Setor de Alimentos do Departamento de Microbiologia do INCQS/Fiocruz. Foram produzidos dois lotes: um utilizando a cepa de E. coli P4328 (EC15/17), e outro lote com a cepa de Klebsiella pneumoniae P4399 (KB03/17). Os dois lotes foram utilizados nos ensaios de “Pesquisa de E. coli em Água” e de “Contagem de Bactérias Mesófilas em Água”. Os lotes foram avaliados quanto à verificação de vácuo, homogeneidade e estabilidade nas temperaturas de referência (≤ -70ºC) e de armazenamento (-20ºC). No teste da homogeneidade, 20 itens de ensaio, de cada um dos lotes, foram separados aleatoriamente e analisados sob condições de repetitividade. Os lotes EC15/17 e KB03/17 foram considerados suficientemente homogêneos e apresentaram concentração final de 4,0 x 102 UFC/mL e 8,0 x 105 UFC/mL, respectivamente. Em relação aos resultados do estudo de estabilidade em longa duração, nas temperaturas de armazenamento e de referência, os itens de ensaio foram considerados estáveis. Concluiu-se que os lotes produzidos apresentaram todas as características esperadas para os ensaios de proficiência. Duas rodadas de EP foram realizadas entre setembro e novembro de 2017.
Abstract
Analytical performance guarantee can be verified through laboratory participation in proficiency tests (PT), which are considered external quality guarantee tools. PT are the use of interlaboratory comparisons for the purpose of evaluating the ability of laboratories to perform a given test or measurement competently and to demonstrate the reliability of the results generated. The objective of this term paper is to produce test items lots in order to be used in two proficiency tests rounds in the counts of mesophilic bacteria and in the research of Escherichia coli in drinking water. The preparation procedures of test items and the control analysis were carried out in the Food Sector of the Microbiology Department of INCQS/Fiocruz. Two batches were produced: one, using the E. coli strain P4328 (EC15/17), and another batch with Klebsiella pneumoniae strain P4399 (KB03/17). For the "Count of Mesophilic Bacteria in Water" and the "E. coli Water Research" assay, both batches produced were used. The lots were evaluated for homogeneity, vacuum check and stability at reference (≤ -70ºC) and storage (-20ºC) temperatures. In the homogeneity test, 20 test items, from each batch, were randomly separated and analyzed under repeatability conditions. Lots EC 15/17 and KB 03/17 were considered sufficiently homogeneous and had a final concentration of 4.0 x 102 CFU/mL and 8.0 x 105 CFU/mL, respectively. In relation to the results of the long-term stability study, at storage and reference temperatures, the test items were considered stable. It was concluded that the produced lots had all the expected characteristics for the proficiency tests. Two rounds were carried out between september and november of 2017.
