A leishmaniose visceral canina representa um importante problema em saúde pública no Brasil. É uma zoonose de transmissão vetorial onde o cão doméstico é a principal fonte de infecção e normalmente o aparecimento dos casos humanos é precedido pelos casos caninos. Com a necessidade de melhoria e diminuir o custo no diagnóstico de LVC, foi introduzido pelo Ministério da Saúde, para este fim, o TR DPP LVC como teste de triagem e EIE como confirmatório. Avaliou-se neste estudo o desempenho do kit de TR DPP LVC produzida por Bio-Manguinhos, através do estudo de estabilidade de longa duração nas temperaturas de 2 a 8 °C e 30 °C durante o prazo de validade do kit de 24 meses e estendeu-se por mais 6 meses, completando 30 meses de estudo nas temperaturas preconizadas. Foram utilizados três lotes nos quais foram avaliados o desempenho do kit (suporte e tampão de corrida) com amostras positiva e negativa. Realizou-se também a avaliação do controle microbiológico e do pH do tampão de corrida de cada tempo de análise de cada lote. Nos lotes analisados até a data de validade e até o 30° mês de análise, a linha teste das amostras positivas se tornaram visíveis no sétimo minuto e a linha controle se tornou visível no sexto minuto. Na avaliação do tampão de corrida quanto ao controle microbiano, foram ausentes em todos os tempos de todos os lotes. Na dosagem do pH dos tampões de corrida, foram observados valores abaixo do especificado em alguns tempos nos lotes avaliados, no entanto estas variações não interferiram no desempenho dos testes. Portanto, até o prazo de validade do kit (24 meses) e até o trigésimo mês de análise os três lotes analisados apresentaram resultados satisfatórios.
Abstract
Canine visceral leishmaniasis is major public health problem in Brazil. It is a vector-borne zoonosis where the domestic dog is the main source of infection and usually the appearance of human cases is preceded by canine cases. With the need to reduce the cost and improve the diagnosis of CVL, was introduced by the Ministry of Health for this purpose, the DPP TR LVC as a screening test and confirmatory EIE. In this study we evaluated the performance of the DPP TR LVC kit produced by Bio-Manguinhos through the long-term stability study at temperatures of 2 to 8 °C and 30 °C during the kit validity period of 24 months and extended for more 6 months, completing 30 months of study at the temperatures recommended. Three batches where we evaluated the performance of the kit (support and running buffer) with positive and negative samples were used. Was also conducted to evaluate the microbiological control of pH and the running time of each analysis of each batch buffer. In batches analyzed until the expiration date and until the 30th month of the test, the test line of the positive samples became visible in the seventh minute and the control line was visible in the sixth minute. In running buffer evaluating the on the microbial control, were absent in all the times of all batches. The measurement of running buffers’ pH was observed values below of the specified in some time in batches reviews, however these changes did not affect the performance of the tests.Therefore, until the expiration date of the kit (24 months) and until the thirtieth month analysis of the three batches analyzed showed satisfactory results.
