Abstract in Portuguese
Os interferons (INFs) são proteínas produzidas por células animais em resposta a infecções virais. Essas substâncias possuem vários efeitos farmacológicos, incluindo antiviral, antitumoral e imunomodulatório. Com o advento da tecnologia do DNA recombinante, vários interferons passaram a ser produzidos e comercializados como Biofármacos. O mais importante é o α2b, que é uma das bases do tratamento das hepatites virais. A única norma oficial que regulamenta a produção de INF-α2b é a Farmacopéia Européia, sendo que restringe-se a matéria prima. O presente trabalho teve como objetivo desenvolver um protocolo para caracterizar a estrutura molecular do INF-α2b em formulações farmacêuticas por espectrometria de massa do tipo MALDI-TOF. Para a análise por MALDI-TOF foi necessário inicialmente desenvolver um método de cromatografia líquida para promover a separação entre o INF-α2b constituinte ativo e minoritário e o soro albumina humana, também componente presente nas formulações farmacêuticas. Primeiramente foi desenvolvido um procedimento baseado em imunoafinidade e cromatografia de gel filtração. As amostras preparadas dessa forma apresentaram homogeneidade protéica por eletroforese, sendo analisadas por dicroísmo circular e fluorescência, o que demonstrou ter ocorrido degradação da estrutura tridimensional. Adicionalmente, essas amostras não geraram espectros de massa. Foi desenvolvido então outro método de separação cromatográfica, baseado em fase reversa, obtendo-se amostras com homogeneidade protéica, revelada por eletroforese. As amostras em solução foram submetidas à digestão com tripsina, levadas ao espectrômetro de massa MALDI-TOF, onde seus fragmentos trípticos foram identificados segundo a correlação entre os resultados encontrados a partir das amostras e do padrão de INF-α2 da Farmacopéia Européia submetido aos mesmos procedimentos. As coberturas de seqüência atingida no padrão foram de 72,7 % e para as amostras de 45,5 %. No produto comercial foi identificada uma ponte disulfeto. Esse trabalho fornece dados importantes que subsidiam o estabelecimento de um protocolo para a análise de INF-α2b em produto final, que poderia substituir o mapa de peptídeos tradicional por cromatografia líquida, com a vantagem de resultar em um maior número de informações sobre a estrutura molecular do biofármaco, importante para a ação farmacológica. Portanto, implementar o controle de qualidade e caracterização estrutural com a introdução de metodologias analíticas sensíveis de maneira que tenhamos informação acerca da segurança, qualidade e eficácia do biofármaco para o devido suporte para as ações de Vigilância Sanitária.
Abstract
Interferons (INFs) are proteins produced by animal cells that respond to viral infections. These compounds exert many biological actions including broad-spectrum antiviral effects, inhibition of tumor cell proliferation and enhancement of immune functions. With the advent of recombinant DNA technology several interferons have been produced and sold as biopharmaceutical. Interferon α2b, is used on the viral hepatitis treatment. The European Pharmacopoeia is the only official regulation that specifies parameters to the INFα2b production, however just for the raw material. The development of analytical protocol by MALDI-TOF mass spectrometry for INFα2b pharmaceutical formulations was the aim of this study. For the MALDI-TOF analyses, initially it was necessary to develop a high pressure liquid chromatography (HPLC) method to separate the minor active component INFα2b from the human serum albumin, also present in the pharmaceutical formulations. Firstly it was developed a procedure based on the immunoaffinity and HPLC-Gel Filtration with protein homogeneity verified by electrophoresis. They were analysed by circular dichroism and fluorescence. However, these samples did not give results when they were submitted to the mass spectrometry. The next step was to develop another separation method using chromatography based on reversed phase. This procedure produced samples with protein homogeneity revealed by electrophoresis. The samples in the solution were submitted to tripsyn digestion, afterwards their triptic fragments were introduced to the MALDI-TOF mass spectrometer and identified by the correlation among the results from the samples and from the European Pharmacopoeia standard INFα2b following the same conditions. The coverage achieved in the standard sequence were 72,7 % and 45,5 % of the samples. In the commercial product a disulfide bridge was identified. This study provides important data that support the establishment of a protocol for the analysis of INFα2b in the final product, which could replace the traditional map of peptides by liquid chromatography with the advantage of resulting in a greater amount of information on the structure molecular of the biopharmaceutical, which is important for the pharmacological action. Therefore, implementing quality control and structural characterization with the introduction of sensitive analytical methodologies in order to obtain information about the safety, quality and efficacy of the biopharmaceutical due to the support for the actions of Sanitary Surveillance.
