Em função de resultados recentes do nosso laboratório, foi observado aumento do número de colônias mielóides totais, independentemente de linhagem, assim como da freqüência de colônias eosinofílicas, formadas em presença de dexametasona, em cultura de células da medula óssea de camundongos. Nos experimentos em que houve adição de dexametasona à cultura foi observado diminuição do número de colônias mielóides totais. Houve aumento de freqüência de colônias eosinofílicas na presença de dexametasona,com alto significado em relação ao grupo controle. A dexametasona afetou a produção de colônias por células hematopoiéticas do sangue de cordão umbilical humano de forma dose-dependente: em concentrações altas, suprimiu a resposta ao GM-CSF, IL-3 E IL-5 sem afetar a porcentagem de colônias eosinofílicas, mostrando um efeito linhagem-inespecífica; em contraste, em concentrações baixas, ela potenciou a formação de colônias eosinofílicas, sem alterar o número total de colônias mielóides, mostrando um efeito linhagem-específico. Nos experimentos em que houve adição de hidrocortisona, observou a diminuição do número de colônias mielóides totais em relação ao grupo controle, com significado estatístico em presença de hidrocortisona. Observou-se um aumento da frequência de colônias eosinofílicas com alto significado estatístico em relação ao grupo controle, na presença de hidrocortisona. Nos experimentos em que houve adição de prostaglandina E2, foi observado diminuição do número de colônias mielóides totais com alto significado estatístico nas concentrações de prostaglandina, mas não houve efeito significativo na concentração. A prostaglandina E2 não teve efeito sobre a frequência de colônias eosinofílicas. Isto indica que a prostaglandina E2 afeta a formação de colônias de células hematopoiéticas de sangue de cordão umbilical humano de forma dose-dependente, mas o efeito é sempre linhagem-inespecífico. O estudo mostrou que tanto a dexametasona como a hidrocortisona aumentaram a freqüência de colônias eosinofílicas. A prostaglandina E2 mostrou efeito inibitório sobre a formação de colônias mielóides de forma dose-dependente, sem efetuar a freqüência de colônias eosinofílicas. Os efeitos são compatíveis com estudos anteriores realizados em medula óssea humana, com a ressalva de uma maior sensibilidade das células de cordão à potenciação pelos glucocorticóides.
