A Doença de Chagas (DC), causada pelo protozoário Trypanosoma cruzi, afeta cerca de 8 - 11 milhões de indivíduos na América Latina. Há 30 anos, o tratamento da DC é feito pelos nitroderivados 2-nitroimidazol (benzonidazol) e 5-nitrofurano (nifurtimox). No entanto, tais drogas apresentam consideráveis efeitos colaterais e baixa eficácia de cura, especialmente na fase crônica da doença. Além disso, a resistência natural e cruzada aos nitroderivados foi descrita para algumas cepas do. parasito, o que aumenta a falha terapêutica e limita as opções de tratamento. A análise proteômica, ideal para o estudo de mudanças globais na expressão de genes em tripanossomatídeos, pode auxiliar na busca de alvos bioquímicos para serem utilizados em novas abordagens quimioterápicas. O presente estudo teve como objetivo identificar proteínas diferencialmente expressas na forma amastigota de duas populações de T. cruzi com resistência selecionada in vivo ao benzonidazol, a população resistente, BZR, e seu par sensível, BZS. Para tal, formas tripomastigotas das populações BZR e BZS de T. cruzi foram mantidas em cultura de células Vero e transformadas em amastigotas após 18h de exposição ao pH 5,0. Os extratos de proteína total obtidos foram separados por eletroforese. bidimensional. Na primeira dimensão, as proteínas foram separadas de acordo com o seu ponto isoelétrico em fitas de gradiente de pH imobilizado de 17cm nas faixas de pH 3-10NL e pH 4-7. Na segunda dimensão, as proteínas foram separadas por peso molecular em gel de poliacrilamida 12% (SDS-PAGE) e corados por Coomasie Blue G-250. As imagens dos géis foram digitalizadas em um densitômetro GS-800 (Bio-Rad) e analisadas através do programa PDQuest 7.3.0 (Bio-Rad). Cerca de 300. e 225 spots dos géis nas faixas de pH 3-10NL e pH 4-7, respectivamente, foram analisados. Os spots selecionados através de análise qualitativa ou quantitativa como diferencialmente ou exclusivamente expressos em uma das populações foram identificados por espectrometria de massa (MS) do tipo Maldi-ToF-ToF. Dos 114 spots submetidos a MS, 107 (94%) foram identificados, correspondendo a 63. proteínas diferentes. Dentre as proteínas identificadas estão aquelas envolvidas com a atenuação dos efeitos gerados pelo metabolismo de drogas, como algumas enzimas da via antioxidante, e outros alvos potenciais de drogas.
