COMPORTAMENTO DOS HEMÓCITOS E DA MATRIZ EXTRACELULAR DE BIOMPHALARIA GLABRATA COM DIFERENTES GRAUS DE RESISTÊNCIA AO SCHISTOSOMA MANSONI: ESTUDO ULTRAESTRUTURAL

O sistema interno de defesa dos moluscos é baseado sobretudo no encapsulamento, fagocitose e destruição das formas invasoras pelos hemócitos. Estudos realizados com o auxílio de técnicas de microscopia eletrônica permitiram elucidar as principais características ultraestruturais destas células e das cápsulas ou complexos encapsulantes por elas formados. Entretanto, as modificações que os hemócitos exibem em animais com diferentes gràus de susceptibilidade ao S. mansoni, o comportamento da matriz extracelular nos complexos encapsulantes e a formação de granulomas ainda não foram suficientemente explorados. Exemplares de B. glabrata provenientes de regiões geográficas distintas foram submetidos às análises histológicas (coloração pela hematoxilina/eosina, sírius-vermelho, orceína e Weigert), ultraestruturais e bioquímicas (técnica de hidroxiprolina). Os achados confirmam que somente um tipo celular (hemócito fagocitico) está presente nas reações celulares. Os elementos da matriz extracelular pesquisados não mostraram participação no interior ou na periferia das reações celulares hemocitárias. As reações teciduais de defesa em B. glabrata contra o S. mansoni são exclusivamente celulares não estando associadas á sintese e deposição de elementos da matriz extracelular.
Resumen en ingles
The internal defense system in snails is mainly based on encapsulation, phagocytosis and destruction of parasite larvae by hemocytes. Electron microscopic studies have demonstrated the principal ultrastructural features of cells, capsule and encapsulating complexes. However, the changes presented by hemocytes in snails with different degrees of susceptibility to S. mansoni, and the role played by the extracellular matrix in granuloma formation (encapsulating complexes) have not yet been sufficiently explored. [MATERIALS AND METHODS]: Normal and S. mansoni- infected B.glabrata from different geographic areas were submitted to histologic (H & E, sirius-red for collagen and orcein stain for elastic tissue), ultrastructural and biochemical methods (hydroxiproline measurement). [RESULTS] Although two cell types were found in hemolymph, in tissue reactions the hemocytes were the only cell-type present. Extracellular components, such as collagen-like and orcein-positive fibers did not participate of the anti-parasite reactions. [CONCLUSIONS]: Tissue reactions against S. mansoni is exclusively dependent on one type of cell: the hemocyte in the snail B. glabrata] synthesis and deposition of extracellular matrix in conspicously absent, in and around zones of reactive cell accumulations.
