A Organização Mundial de Saúde estima que 80% da população mundial depende da medicina tradicional, onde cerca de 25.000 espécies de plantas são usadas nas preparações de extratos vegetais. No semi-árido brasileiro, que inclui oito estados e corresponde a 11,5% do território nacional, estima-se haver 8 mil espécies vegetais pouco estudadas quanto às atividades farmacológicas. Diante dessa vasta biodiversidade e da necessidade da descoberta de novas moléculas bioativas, é de fundamental importância o estudo farmacológico da flora dessa região, tendo em vista a acelerada degradação ambiental que põe em risco de extinção um grande número de espécies. Este trabalho teve como objetivo a investigação da atividade anti-Leishmania amazonensis, anti-Trypanosoma cruzi e imunomoduladora em extratos de plantas nativas ou endêmicas do semi-árido brasileiro. O material vegetal utilizado neste estudo foi coletado e identificado por uma equipe de botânicos integrantes do Instituto do Milênio do Semi-Árido (IMSEAR/MCT). Os extratos foram obtidos a partir de 69 espécies vegetais e com solventes de diferentes polaridades químicas. Para os ensaios de atividade farmacológica, testaram-se inicialmente os extratos para a determinação de concentrações tóxicas para células de camundongos (esplenócitos), sendo utilizadas nos ensaios subseqüentes aquelas com até 30% de citotoxicidade. Para os ensaios de atividade antiparasitária in vitro, os parasitas foram cultivados em presença dos extratos e o percentual de inibição foi medido através da determinação do metabolismo do MTT. Os ensaios de inibição da linfoproliferação foram realizados cultivando as células na presença do mitógeno concanavalina A e dos extratos. A proliferação foi determinada através da medida da incorporação de timidina tritiada. Para a avaliação da inibição da produção de óxido nítrico (NO), cultivou-se macrófagos peritoneais de camundongo na presença de IFN-y e LPS e dos extratos. A produção de NO foi avaliada pela dosagem de nitrito, através do método de Griess. Dos 174 extratos avaliados, 3 apresentaram atividade anti-leishmania acima de 90% e 10 apresentaram atividade anti-T cruzi superior a 60%. Em relação à atividade imunomoduladora, 47 extratos apresentaram atividade inibidora da linfoproliferação superior a 80%, enquanto que 16 dos 174 extratos testados apresentaram atividade de inibição da produção de NO acima de 70%. Frações obtidas a partir de 2 extratos de vegetais com atividade imunomoduladora foram reavaliadas, identificando-se as ativas. Os extratos e frações avaliados apresentam um potencial farmacológico importante, sobretudo em relação à atividade imunomoduladora. Outros extratos ativos serão fracionados e estas, assim como as ftações já obtidas serão re-fracionadas para reavaliação com o intuito identificar as moléculas responsáveis pela atividade, podendo ser no futuro candidatas a novos fármacos.

The World Health Organization estimates that 80% of the world's population depends on conventional medicine, in which around 25.000 plant species are used in the preparation of extracts. In the BraziUan semi-arid, which includes eight states and corresponds to 11,5% of the national territory, it is estimated that there are 8 thousand plant species scarcely studied regarding its pharmacological activities. Considering this vast biodiversity and the need for the discovery of new bioactive molecules, the pharmacological study of this regions’ flora is of fundamental importance, given the accelerated enviroimiental degradation that puts in risk of extinction a great number of species. In this work we aimed at the investigation of mti-Leishmania amazonensis, anti- Trypanosoma cruzi and immuno-modulatory activities of extracts of native or endemic plants from the Brazilian semi-arid. The vegetal material used in this study was collected and identified by a team of botanists who were part of the Institute of the Milennium of the Semi-Arid (MCT). The extracts were obtained fi-om 69 plant species with solvents of different chemical polarities. For the assays of the pharmacological activity, the extracts were initially tested to determine the toxic concentrations for mammal cells (mouse splenocytes), being used in subsequent assays those with up to 30% cytotoxicity. For the assays of antiparasitic activities in vitro, the parasites were cultivated in the presence of the extracts and the percentage of inhibition was measured through the determination of the metabolism of the MTT. The lymphoproliferation inhibition assays were carried out by cultivating the cells in the presence of the mitogen concanavalin A and of the extracts. The proliferation was determined through the measurement of ^H-thymidine incorporation. For the evaluation of the inhibition of nitric oxide (NO) production, mouse peritoneal macrophages were cultivated in the presence of IFN-7 and LPS and the extracts. The production of NO was evaluated by the dosage of nitrite, through the Griess method. Of the 174 extracts evaluated, 3 showed anti-leishmania activity above 90% and 10 showed anti- T. cruzi activity superior to 60%. Regarding the immunomodulatory activity, 47 extracts showed inhibitory activity of lymphoproliferation superior to 80%, whereas 16 out of 174 extracts tested showed 70% of inhibitory activity on NO production. Fractions obtained from 2 extracts with immunomodulatory activity were tested afterwards to identify the active ones. The extracts and fractions evaluated showed an important pharmacological potential, especially in relation to the immunomodulatory activity. Other active extracts will be fi*actioned and these, as well as the already obtained fractions will be re-fi'actioned for re-evaluation, with the purpose of identifying the molecules responsible for the activity, putative candidates to new pharmaceutical drugs.
