A tuberculose é uma doença de importância mundial, causando, ainda hoje, grandes taxas de mortalidade e morbidade na população mundial. O reconhecimento dos padrões moleculares associados a patógenos pelas células do sistema imune durante a infecção é essencial para a ativação da resposta imune inata. Os macrófagos são primeira linha de defesa no pulmão, sítio de infecção mais comum desta doença, possuem diversos tipos de receptores responsivos aos padrões moleculares associados a patógenos (PAMPs). A família de receptores do tipo NOD (NLR) é importante sinalizadora para desencadear a montagem do inflamassoma, uma plataforma molecular capaz de processar citocinas inflamatórias para sua forma ativa, principalmente IL-1\03B2 e IL-18. Ainda na frente de resposta inata, há os corpúsculos lipídicos (CL), organelas citoplasmáticas dinâmicas capazes de aumentar em tamanho e número frente a infecções que participam da regulação da produção de mediadores inflamatórios e levam ao favorecimento da sobrevivência dos patógenos intracelulares O presente estudo visa, portanto, investigar a possível relação entre as proteínas componentes do inflamassoma e os CLs durante a resposta de macrófagos à infecção por Mycobacterium bovis, questão que permanece por ser melhor elucidada. Para tal, macrófagos derivados de medula óssea de camundongos machos C57 Black 6 foram infectados com M. bovis BCG por 24 horas. Posteriormente as células foram analisadas por microscopia de fulorescência e confocal, objetivando verificar se haveria presença das componentes do inflamassoma nos CLs. Os resultados demonstram que, NALP3 e caspase-1 estariam co-localizados com os CLs, mas não ASC. A inibição de biogênese de CLs mediada pela droga C75, inibidora da enzima ácido graxo sintase, reduziu a secreção de IL1\03B2, principal substrato clivado pelo inflamassoma NLRP3. Através da análise por WB, nossos resultados demonstraram um efeito inibitório do C75 tanto na expressão quanto no processamento de IL-1\03B2, sugerindo um papel para os corpúsculos lipídicos na ativação do inflamassoma NLRP3.

Tuberculosis is a disease of worldwide importance, causing great mortality and morbidity rates in the world population. Recognition of molecular patterns associated with pathogens by cells of the immune system during infection is essential for the activation of the innate immune response. Macrophages are the first line of defense in the lung, the most common site of infection of this disease, and have several types of receptors responsive to molecular patterns associated with pathogens (PAMPs). The NOD receptor family (NLR) is an important marker for triggering the assembly of inflammassome, a molecular platform of processing inflammatory cytokines into their active form, mainly IL-1\03B2 and IL-18. Still in the front of innate response, there are lipid droptels (LD), dynamic cytoplasmic organelles of increasing in size and number against infections that participate in the regulation of the production of inflammatory mediators and lead to favoring the survival of intracellular pathogens The present study therefore aims to investigate the possible relationship between the proteins components of the inflammassome and LDs during the response of macrophages to Mycobacterium bovis infection, a question that remains to be elucidated. To that end, bone marrow derived macrophages of C57 Black 6 male mice were infected with Mycobacterium bovis BCG for 24 hours. Afterwards the cells were analyzed by fulcrum and confocal microscopy, to verify if there was presence of the components of the inflammassoma in the LDs. The results demonstrate that, NALP3 and caspase-1 would be co-localized with the LDs, but not ASC. Inhibition of CL biogenesis mediated by the enzyme fatty acid synthase inhibitor C75 reduced secretion of IL-1\03B2, the major substrate cleaved by the inflammatory neoplasm NLRP3. Through WB analysis, our results demonstrated an inhibitory effect of C75 on both IL-1\03B2 expression and processing, suggesting a role for lipid droplets in the activation of inflammome NLRP3.
