O uso contínuo de inseticidas piretroides (PI) no contexto do Programa Nacional de Controle
da Dengue tem levado à seleção de populações de Aedes aegypti resistentes à maioria desses
compostos. O presente estudo caracterizou a susceptibilidade de populações pernambucanas
de A. aegypti aos PI e identificou os possíveis mecanismos envolvidos na resistência.
Amostras do mosquito dos municípios de Águas Belas (AB), Arcoverde (AV), Caruaru (CA)
e da Ilha de Fernando de Noronha (FN) foram analisadas. Fêmeas foram expostas a papéis
impregnados com cipermetrina, deltametrina ou lambda-cialotrina. A linhagem Rockefeller
foi usada como controle dos ensaios. A razão de resistência (RR) foi estimada pela
mortalidade das fêmeas e o tempo nocaute. Fêmeas resistentes e susceptíveis foram analisadas
por microscopia eletrônica de varredura para estimar a espessura cuticular e por PCR para
detectar mutações Kdr no gene Nav. Testes bioquímicos foram feitos para quantificar as
atividades de enzimas GSTs e α-esterases. Os resultados mostram que, com exceção de FN, as
demais populações são resistentes aos PI e exibem diferentes valores de RR para os três
compostos. Valores mais altos foram observados para cipermetrina em AV e CA e para -
cialotrina em AV. Fêmeas resistentes apresentaram a espessura da cutícula significativamente
maior que as susceptíveis, caracterizando o envolvimento do mecanismo de penetração
reduzida. Mutações nos sítios 1011, 1016 e 1534 do gene Nav foram detectadas em todas as
populações e associadas com a resistência em AB, AV e CA. Os padrões enzimáticos estavam
mais alterados nas populações resistentes. Este trabalho caracterizou pela primeira vez o
mecanismo de penetração reduzida em A. aegypti, e evidenciou a baixa efetividade do uso dos
PI para o controle local de A. aegypti. Este mecanismo até então pouco investigado pode estar
difundido em populações naturais de vetor, comprometendo o sucesso de programas de
controle e, portanto, merece maior investigação.
Resumen en ingles
The continuous use of pyrethroids (PY) in the context of the National Dengue Control
Program has triggered intense selection pressure for these compounds and the consequent
emergence of resistant populations. This project characterized the susceptibility of A. aegypti
populations from Pernambuco to PY and identified possible mechanisms involved with
resistance. Mosquito samples from the municipalities of Águas Belas (AB), Arcoverde (AV),
Caruaru (CA) and Fernando de Noronha Island (FN) were analyzed. Females were exposed to
papers impregnated with cypermethrin, deltamethrin or lambda-cyhalothrin. The Rockefeller
strain was used as a control test. The resistance ratio (RR) was estimated by female mortality
and knockdown time. Resistant and susceptible females were analyzed by scanning electron
microscopy to estimate cuticle thickness and by PCR to detect Kdr mutations in the Nav gene.
Biochemical tests were performed to quantify the activities of GSTs and α-esterases. The
results show that, excepting FN, all populations are resistant to PY and have different RR
values for the three compounds. Higher values were observed for cypermethrin in AV and CA
and for lambda-cyhalothrin in AV. Resistant females had cuticle thickness significantly
thicker than susceptible females, characterizing the involvement of the reduced penetration
mechanism. The Ile1011Met, Val1016Ile and Phe1534Cys mutations were detected in Nav
gene in all populations and associated with resistance in AB, AV and CA. Enzyme standards
were more altered in resistant populations. This work caracterizes for the first time the
mechanism of penetration reduced in A. aegypti, and evidences the ineffectiveness of the use
of PY for the local control of A. aegypti. This mechanism, hitherto little investigated, may be
widespread in natural populations of vector, compromising the success of control programs
and therefore deserves further investigation.
