Hidridoma é uma linhagem celular produzida em laboratório a partir da fusão de células linfóides produtoras de anticorpo com células cancerígenas não produtoras. São capazes de se proliferar e produzir anticorpos monoclonais específicos de forma contínua. Anticorpos monoclonais são glicoproteínas de alto valor agregado, indicados para uso terapêutico e diagnóstico, e seu uso tem sido proposto para tratamento de infecções bacterianas resistentes a múltiplas drogas, tais como Staphylococcus aureus Resistente à Meticilina (MRSA), que atualmente são responsáveis por causar 29% das infecções hospitalares no Brasil. O objetivo deste trabalho foi investigar os fatores que podem influenciar na obtenção de anticorpos monoclonais em cultura de células de hibridoma murino anti-PBP2a, em frasco tipo spinner, secretoras de anticorpo monoclonal anti-PBP2a, que confere resistência aos Staphylococcus aureus. Os efeitos da variação da concentração do inóculo celular inicial e da velocidade de agitação sobre o crescimento celular e a obtenção de anticorpo monoclonal anti-PBP2a de MRSA (Mab-PBP2a) foram avaliados através do planejamento fatorial completo 22, enquanto a taxa de produção de anticorpo monoclonal específico foi avaliada por um plano fatorial completo 23 Foi verificada uma tendência de aumento da concentração de Mab-PBP2a com o uso de maior concentração do inóculo celular inicial e velocidade de agitação e uma redução na taxa específica de produção anticorpo monoclonal anti-PBP2a de MRSA foi verificada. A velocidade de agitação foi a variável que apresentou maior influência sobre a concentração máxima de células, secreção de Mab-PBP2ae taxa específica de produção de anticorpo monoclonal. Quando as células de hibridoma secretoras do anticorpo monoclonal anti-PBP2a de MRSA foram cultivadas em um frasco do tipo spinner com eixo fixo agitação e concentração celular inicial de 1,3 x105células viáveis/mL a 112rpm, não foi observado efeito negativo sobre a concentração do Mab-PBP2a, ao final do segundo dia de cultivo, sendo obtida uma concentração de 34\03BCg/mL. Foi observado que a concentração do inóculo celular inicial pode ser reduzida, devendo ser considerada a concentração de anticorpo monoclonal secretado e um adequado cultivo celular de hibridomas sem prejuízos na secreção do anticorpo monoclonal anti-PBP2a de MRSA.

Hybridoma is a cell line produced in laboratory from the fusion of an antibody-producing lymphocyte and a nonantibody-producing cancer cell. They are able to proliferate and produce a continuous supply of a specific monoclonal antibody. Monoclonal antibodies are glycoproteins with high value-added and are applied in diagnostic and therapeutic, where its use has been proposed to the treatment of multidrug-resistant infections like Methicillin Resistant Staphylococcus aureus (MRSA), which currently account for 29% of nosocomial infections in Brazil. The aim of this work was to investigate in a spinner flask, the factors that may influence the attainment of monoclonal antibodies in cultured murine hybridoma cells that secrete the monoclonal antibody anti-PBP2a, which confers resistance to Staphylococcus aureus. The effects on cell growth and secretion of monoclonal anti-PBP2a of MRSA (Mab- PBP2a) were verified by varying the initial cell inoculum size and agitation speed, and it was assessed using full factorial design 22, while the rate of production of specific monoclonal antibody was evaluated by a full factorial design 23 It was observed a tendency of increased concentration of Mab-PBP2a when using a higher initial cell inoculum size and agitation speed, while a reduction in the rate specific production of Mab-PBP2a was verified. The stirring speed was the variable factor that had the most important influence on the maximum cell concentration, secretion of Mab-PBP2a and specific rate of production of monoclonal antibody. When MRSA-secreting hybridoma cells were grown in a spinner flask type with fixed stirring shaft at a initial cell concentration of 1,3x105viable cell/mL and 112rpm, no negative effect on the Mab-PBP2a concentration 34\03BCg/mL obtained at the end of the second day of cultivation was observed. It was observed that the initial cell inoculum size can be reduced, if considered the concentration of secreted monoclonal antibody and a suitable hybridoma cell culture without reducing the secretion quality of monoclonal anti-MRSA PBP2a.
