OBTENÇÃO E CARACTERIZAÇÃO DE FRAGMENTOS F(AB')2 OBTIDOS A PARTIR DE ANTICORPOS MONOCLONAIS MURINOS ANTI-PBP2A DE STAPHYLOCOCCUS AUREUS RESISTENTES À METICILINA (MRSA)

As infecções causadas por MRSA são um problema de saúde mundial, especialmente devido à dificuldade de tratamento, alto grau de virulência e elevada morbidade associada. As cepas de MRSA caracterizam-se por possuir uma proteína de baixíssima afinidade por \03B2-lactâmicos, a PBP2a. Por ser uma bactéria multirresistente, diversas alternativas de tratamento têm sido pesquisadas, como coquetéis de antimicrobianos e imunoterapias, porém até agora nenhuma obteve sucesso. Desta vez, tendo em vista a PBP2a como um alvo em potencial, encontrase em desenvolvimento um anticorpo monoclonal anti-PBP2a de MRSA. Fragmentos de anticorpos, contendo apenas as duas porções Fab\2019, são amplamente descritos na literatura como ferramentas imunoquímicas, reagentes de diagnóstico e terapêutica, graças à sua rápida farmacocinética e baixa imunogenicidade Os fragmentos F(ab)2\2019 foram obtidos por digestão enzimática com papaína e pepsina e purificados através de cromatografia de afinidade utilizando resinas de Proteína A, como a MabSelectSure (GE) e concentradas por unidades filtrantes Amicon® MWCO 50 kDa. Suas afinidades pela PBP2a foram testadas através de um ensaio ELISA do tipo indireto e um Western Blot. Através de um ensaio de neutralização bacteriana, verificou-se a atividade neutralizante dos fragmentos após a digestão. Foi feito também um ensaio de farmacocinética com camundongos. Os resultados indicaram que foi possível a obtenção e o isolamento de fragmentos F(ab\2019)2 por digestão com papaína e pepsina, sendo que a pepsina apresentou melhor produção de F(ab\2019)2 com melhor rendimento. O ELISA e o Blot demonstraram que os F(ab\2019)2 não perderam afinidade pela PBP2a, mesmo após o processo de digestão enzimática, assim como não perderam sua capacidade neutralizante, obtendo resultados próximos aos apresentados pelo anticorpo não digerido. E como esperado, nos ensaios de farmacocinética o F(ab\2019)2 apresentou eliminação mais rápida (entre 12 e 18 horas) se comparado à IgG (aproximadamente 9 dias).

Infections caused by MRSA are a worldwide health problem, especially due to the difficulty of treatment, high degree of virulence and high associated morbidity. MRSA strains are characterized by having a protein with very low affinity for \03B2-lactams, the PBP2a. As a multiresistant bacteria, various alternatives for treatment have been researched, such as cocktails of antibiotics and immunotherapies, but so far none has succeeded. This time, looking up to PBP2a as a potential target, it is in development a monoclonal antibody anti-MRSA PBP2a. Antibody fragments containing only the two Fab\2019 portions, are widely described in the literature as immunochemical tools, reagents for diagnostics and therapeutics, especially due to its low immunogenicity and rapid pharmacokinetics. The F(ab\2019)2 fragments were obtained by enzymatic digestion with papain and pepsin and purified by affinity chromatography using Protein A resins such as MabSelectSure (GE) and concentrated by Amicon® filtration units MWCO 50 kDa. Their affinity for PBP2a was tested with an ELISA-type indirect and a Western Blot essay. Through a bacterial neutralization assay, it was evaluated the neutralizing activity of the fragments after digestion. It was also done pharmacokinetics test in mice. The results indicated that it was possible to obtain the isolation of F(ab\2019)2 fragments by digestion with pepsin and papain, and pepsin showed improved production of F(ab\2019)2 with better performance. ELISA and Blot demonstrated that F(ab\2019)2 affinity for PBP2a was not lost even after the enzymatic digestion process, nor was lost its neutralizing capacity, obtaining results similar to those presented by the undigested antibody. As expected, in the pharmacokinetics tests, F(ab\2019)2 has presented a faster elimination (between 12 and 18 hours) if compared to IgG (approximately 9 days).
