EFEITO IN VITRO DE DIAMINAS, AZLACTONAS E OUTROS DERIVADOS QUÍMICOS SOBRE TRYPANOSOMA CRUZI

Nesta tese foram avaliados 81 novos compostos sintéticos contra epimastigotas de Trypanosoma cruzi Dm28c pelo método MTT, sendo 4 mais eficazes em epimastigotas: GSC-01 (Indice de Seletividade IS=17,86), GIB-24 (IS=16,4), CGE (IS=8,39) e EPA-35 (IS=6,74). O composto EPA-35 foi o mais ativo contra formas amastigotas intracelulares (IS=95,04). A azlactona EPA-35 e a diamina GIB-24 foram utilizados em estudos adicionais. Com EPA-35 observou-se por citometria que a membrana plasmática de epimastigotas tratados (15 a 50 μM) manteve-se inalterada, ocorrendo, porém, alteração no potencial de membrana mitocondrial. Redução de tamanho e arredondamento foram observados por microscopia eletrônica de varredura (MEV) após tratamento de epimastigotas com EPA-35 a 33 ou 50 μM. Amastigotas intracelulares tratados mostraram citoplasma mais eletrondensos por microscopia eletrônica de transmissão (MET), com perda da estrutura celular normal. No tratamento com GIB-24 observou-se que o tratamento de células Vero infectadas não impediu que amastigotas intracelulares completassem o ciclo intracelular, havendo porém diminuição no número de parasitas liberados com aumento na razão %amastigotas/%tripomastigotas. Significativa dissipação do potencial de membrana mitocondrial ocorreu com 6 horas de tratamento, previamente à permeabilização da membrana plasmática. Epimastigotas arredondados, com redução do tamanho celular e vacuolização do citoplasma foram observados por MEV e MET. As alterações induzidas por GIB-24 sugerem processo de morte incidental. Houve parada no ciclo celular de epimastigotas após 3 horas de tratamento (6 e 12 μM), ficando a maioria dos epimastigotas na fase G1. Análise proteômica inicial de epimastigotas resistentes a 15 μM GIB-24 indicou que o composto age principalmente sobre componentes mitocondriais envolvidos no ciclo de Krebs e na manutenção da homeostase oxidativa dos parasitas. Nossos dados sugerem que EPA-35 e GIB-24 são ativos contra T. cruzi merecendo avaliações posteriores.

In this thesis, 81 new synthetic compounds were tested against epimastigotes of Trypanosoma cruzi Dm28c by the MTT method, with 4 being more effective in epimastigotes: GSC-01 (IS = 17.86), GIB-24 (IS = 16.4), CGE (IS = 8.39) and EPA-35 (IS = 6.74). EPA-35 was the most active against intracellular amastigote forms (SI = 95.04). EPA-35 azlactone and the diamine GIB-24 were further evaluated. With EPA-35, it was observed by cytometry that the plasma membrane of treated epimastigotes (15 to 50 μM) remained unchanged, but there was alteration in mitochondrial membrane potential. Reduction in size and rounding of epimastigotes were observed by scanning electron microscopy (SEM) after treatment with EPA-35 at 33 or 50 μM. Treated intracellular amastigotes showed more electron-emitting cytoplasm by transmission electron microscopy (TEM), with loss of normal cell structure. In the treatment with GIB-24, it was observed that treatment of infected Vero cells did not prevent intracellular amastigotes completing the intracellular cycle, but there was a decrease in the number of parasites released with an increase in the % amastigotes /% trypomastigote ratio. Significant dissipation of the mitochondrial membrane occurred with 6 hours of treatment, prior to permeabilization of the plasma membrane. Rounded epimastigotes with cell size reduction and cytoplasmic vacuolation were observed by SEM and TEM. Changes induced by GIB-24 suggest an incidental death process. The epimastigote cell cycle was stopped after 3 hours of treatment (6 and 12 μM), with the majority of epimastigotes remaining in the G1 phase. Initial proteomic analysis of 15 μM resistant epimastigotes GIB-24 indicated that the compound acts mainly on mitochondrial components involved in the Krebs cycle and in maintaining the oxidative homeostasis of the parasites. Our data suggest that EPA-35 and GIB-24 are active against T. cruzi deserving further evaluation.
