Mosquitos do gênero Aedes são implicados na transmissão de várias doenças infecciosas. Aspectos como dinâmica populacional, estrutura e diversidade genética de populações de mosquitos são importantes para auxiliar no desenvolvimento de ferramentas mais eficientes de monitoramento e controle vetorial. O presente estudo teve como objetivo principal avaliar a estrutura genética das espécies A. aegypti e A. albopictus em São Lourenço da Mata (SLM). As coletas de ovos foram realizadas em três áreas: Arena Pernambuco (AP); Obra de Maria (OM) e Parque Capibaribe (PC), e ocorreram em dois períodos distintos, separados por um ano. As larvas foram mantidas no insetário até a emergência dos insetos adultos e estes foram identificados por taxonomia clássica. Em seguida foram submetidos individualmente à extração de DNA, PCR para amplificar fragmentos dos genes mitocondriais COI e ND5 e sequenciamento. As coletas revelaram maior abundância da espécie A. albopictus nas áreas de AP e OM, e prevalência de A. aegypti em PC. A análise do número de haplótipos e diversidade nucleotídica (π) de COI para A. albopictus revelou uma diminuição desses índices ao longo dos dois anos do estudo. Por outro lado, o número de haplótipos e π de ND5 em SLM não sofreram variação entre os dois anos. Resultados similares foram observados para COI em A. aegypti, com exceção do número de haplótipos e π de ND5 que aumentaram em um ano. Os testes de neutralidade demonstram que, em geral, há uma expansão populacional do gênero Aedes em SLM, cujos marcadores não estão sob efeito de seleção. Os dados de índice de fixação (FST) para A. albopictus indicaram diminuição significativa da diferenciação genética na comparação entre os dois anos. Enquanto que os dados de FST para A. aegypti demonstraram moderada divergência. Os resultados obtidos sugerem que as populações de A. albopictus e A. aegypti de SLM apresentam atualmente baixa diferenciação genética, quando comparadas ao período de coleta anterior.

Mosquitoes of the Aedes genus are implicated in the transmission of various infectious diseases. Aspects such as population dynamics, structure and genetic diversity of mosquito populations are important to assist the development of more efficient monitoring and vector control tools. This study had as the main goal to evaluate the genetic structure of A. aegypti and A. albopictus species in S. Lourenço da Mata (SLM). The egg collections were done in three areas: Pernambuco Arena (PA); Obra de Maria (OM) and Capibaribe Park (CP), and occurred in two distinct periods, separated by a year. The larvae were kept in the cage until surfaced the adult insects and these were identified by classical taxonomy. Then they were individually subjected to DNA extraction, PCR to amplify fragments of the mitochondrial genes COI and ND5 and sequencing. The collections showed greater abundance of the A. albopictus species in the PA and OM areas, and supremacy of A. aegypti in CP. The analysis of the number of haplotypes and nucleotide diversity (π) of COI for A. albopictus revealed a decrease in these indexes all over the two years of this review. On the other hand, the number of haplotypes and π of ND5 in SLM did not change in two years. Similar results were observed for COI in A. aegypti, except for the number of haplotypes and π of ND5 that increased in one year. The neutrality tests show that, in general, there is a population expansion of the Aedes genus in SLM, whose markers are not under selection effect. Fixed index (FST) data for A. albopictus indicated a significant decrease in genetic differentiation in the two years proportion. While the FST data for A. aegypti showed moderate divergence. The results obtained suggest that the A. albopictus and A. aegypti populations of SLM introduce low genetic differentiation when compared to the previous collection period.
