A leptospirose é uma zoonose relevante para a saúde pública, afetando mais de
um milhão de casos a cada ano no mundo. As leptospiras colonizam os rins de
animais reservatórios e são excretadas vivas no ambiente, principalmente pela
urina. O ambiente é a principal forma de transmissão indireta. O objetivo deste
trabalho foi isolar bactérias do gênero Leptospira em amostras ambientais (esgoto
e solo) de uma área urbana endêmica, classificando o sorogrupo representante.
Foram coletadas 10 amostras (6 de esgoto e 4 de solo) no bairro de Pau da Lima,
Salvador-Ba, em abril de 2016. Em laboratório, as amostras foram filtradas com
filtro de seringa de 0,22 μm e adicionadas em meio de cultura EMJH, contendo
100 μg/mL, 200 μg/mL e 700 μg/mL do antibiótico 5-Fluorouracil (5-FU). As
amostras foram incubadas em estufa a 29°C durante dois meses. A observação
das culturas foi feita através do microscópio de campo escuro, com objetiva de
20x. Foi realizado o isolamento das culturas de bactérias com morfologia
helicoidal, compatíveis com o gênero Leptospira, em placa com meio EMJH sólido
até obtenção de uma cultura pura. As culturas isoladas de Leptospira foram
submetidas à extração de DNA e em seguida ao seqüenciamento genético, com
base na análise filogenética de rRNA 16S. As sequências foram corrigidas usando
BioEdit 7.2.5 (Ibis Biosciences) e comparadas utilizando BLAST com as
sequências disponíveis no NCBI. Foram identificadas as bactérias L. kmetyi
(patogênica) e L. idonii (saprofítica) nas amostras de solo. Já nas amostras de
esgoto, foram encontradas L. idonii e L. yanagawae, ambas saprofíticas. As três
concentrações do 5-FU foram semelhantemente eficazes na inibição dos
contaminantes. As amostras ambientais do Bairro de Pau da Lima foram positivas
para Leptospira saprofíticas e patogênicas, ressaltando a importância da
caracterização ambiental das bactérias deste gênero para a compreensão dos
riscos de transmissão aos humanos e animais.
