Abstract
Casearia sylvestris (Salicaceae) is found in South America and presents antiulcerogenic, cytotoxic, antimicrobial, anti-inflammatory and antihypertensive activities. Aim ofthestudy: Toassessthe in vivoandexvivo antitumor actionofafractionwithcasearins(FC)andits
main component-CasearinX-isolatedfrom C. sylvestris leaves.
Materials andmethods: Firstly,Sarcoma180bearingSwissmiceweretreatedwithFCandCasXfor
7 days.Secondly,BALB/cnudeanimalsreceivedhollow fibers withcoloncarcinoma(HCT-116)orglio-
blastoma (SF-295)cellsandweretreatedwithFCfor4days.On5thday,proliferationwasdeterminedby
MTT assay.
Results: FC 10and25mg/kg/dayi.p.and50mg/kg/dayoralandCasX25mg/kg/dayi.p.and50mg/kg/
day oralrevealedtumorgrowthinhibitionratesof35.8,86.2,53.7,90.0and65.5%andsuchtumors
demonstratedraremitosesandcoagulationnecrosisareas.Similarly,FCreducedmultiplyingofHCT-116
and SF-295cellswhenevaluatedbytheHollowFiberAssay(2.5and5mg/kg/dayi.p.and25and50mg/
kg/day oral),withcellgrowthinhibitionratesrangingfrom33.3to67.4%(po0.05). Flowcytometry
experimentsrevealedthatFCreducedmembraneintegrityandinducedDNAfragmentationandmi-
tochondrial depolarization(po0.05).
Conclusions: FC andCasXwereefficient antitumorsubstancesagainstmurineandhumancancercells
and causedreversiblemorphologicalchangesinliver,kidneysandspleens,emphasizingclerodanedi-
terpenes asanemergingclassofanticancermolecules.
