A Febre Amarela (FA) constitui um grave problema de saúde pública mundial, principalmente nos países tropicais (embora este cenário venha sendo modificado pois o clima continua sendo afetado pelo aquecimento global). Apesar da eficiência comprovada da vacina convencional atenuada (17DD), existem registros de casos adversos graves pós-vacinação, incluindo o registro de óbitos. Adicionalmente a vacina 17DD não é recomendada para infantes, mulheres grávidas, pessoas com imunodeficiência e aquelas que apresentam alguma alergia aos componentes do ovo. Considerando todos estes fatores, faz-se necessário o desenvolvimento de estratégias de vacinação ainda mais seguras contra a FA, como por exemplo vacinas de DNA. As vacinas genéticas podem ser mais facilmente manipuladas e dosadas, não necessitam de baixas temperaturas para o acondicionamento/distribuição e não são capazes desencadear efeitos adversos graves por não se tratarem de agentes infecciosos. Neste trabalho, nós descrevemos o desenvolvimento e avaliação da expressão de antígenos codificados por vacinas de DNA contra o Vírus da Febre Amarela (VFA). O genoma do VFA codifica três genes para proteínas estruturais (Capsídeo, Membrana e Envelope - E) e sete genes que codificam proteínas não-estruturais. Considerando que a proteína E é reconhecida como a principal proteína da superfície viral, e principal alvo para a indução da produção de anticorpos neutralizantes, nossas formulações vacinais foram baseadas neste antígeno. O antígeno vacinal foi otimizado para a expressão em células eucarióticas e subclonado em um vetor eucariótico de expressão, gerando a construção p/YFEOPT . Uma segunda construção foi ainda obtida, pL/YFEOPT, que codifica o antígeno vacinal fusionado à região C-terminal da Proteína de Associação à Membrana Lisossomal humana - hLAMP (visando desviar a apresentação antigênica para a via do Complexo Maior de Histocompatibilidade de classe II - MHCII). Estas construções foram utilizadas para transfectar células eucarióticas e a expressão, dos antígenos codificados, foi avaliada através de ensaios de imunofluorescência e western-blot. A próxima etapa deste trabalho será a avaliação do efeito de proteção da vacina de DNA pL/YFEOPT em modelos primatas não-humanos. De acordo com os resultados obtidos, pretendemos avançar para ensaios clínicos em humanos
