O câncer de mama triplo negativo é conhecido como o subtipo molecular de pior prognóstico quando comparado aos demais. Estudos atuais demonstram que a matriz extracelular (MEC) é um fator importante para o caráter agressivo de alguns tipos de câncer. Dentre os elementos da MEC, os proteoglicanos são essenciais para a manutenção da estrutura do tecido, como por exemplo o versican (VCAN). Esse proteoglicano, quando fragmentando, gera proteólitos que têm sido relacionados à atividade de matrikinas, uma vez que apresentam função imunomoduladora associada à progressão tumoral e a angiogênese. Todavia, a relação de matrikinas com o remodelamento da matriz ainda não foi elucidada. Objetiva-se avaliar a relação entre as versicanases, microambiente tumoral inflamatório e desenvolvimento de metástases em modelo murino 4T1. Foram realizadas análises histomorfológicas em amostras de tumor primário, linfonodo regional e pulmão nos grupos de 7, 14, 21 e 28 dias de crescimento tumoral e análises da expressão imuno-histoquímica de VCAN, VKINA, as desintegrinas e metaloproteinases com domínios trombospondinas (ADAMTS) -1, -5 e -15, bem como análise do perfil imunofenotípico de células da resposta imune em sangue periférico e infiltrado tumoral. Ao longo dos 28 dias, houve um aumento crescente da expressão de VCAN. Contudo, a expressão de VKINA atingiu maior mediana aos 21 dias de crescimento tumoral, assim como a ADAMTS15. ADAMTS1 e 5 foram mais expressas em estroma aos 21 e 28 dias de inoculação tumoral. VCAN, ADAMTS 1 e 5 apresentaram maior expressão nas últimas semanas do experimento, com correlação direta ao perfil predominantemente pró-inflamatório. No entanto, VKINA e ADAMTS15 apresentaram maior expressão na terceira semana, quando há o surgimento de metástases nesse modelo Além da alta expressão de VKINA em 21 dias, esse protéolito teve relação com perfil imune predominantemente anti-inflamatório, criando um microambiente favorável a progressão do tumor. Os resultados sugerem uma relação entre degradação da MEC e perfil inflamatório. Contudo, ainda se faz necessário compreender os efeitos destes perfis imunes no remodelamento da matriz extracelular com relação das versicanases e seu proteólito nos diferentes tempos de progressão tumoral
Abstract
Triple negative breast cancer is known as the molecular subtype with the worst prognosis when compared to the others. Current studies demonstrate that the extracellular matrix (ECM) is an important factor in the aggressive nature of some types of cancer. Among the elements of the ECM, proteoglycans are essential for the maintenance of tissue structure, such as versican (VCAN). This proteoglycan, when fragmented, generates proteolites that have been related to the activity of matrikins, since they have an immunomodulatory function associated with tumor progression and angiogenesis. However, the relationship between matrikins and matrix remodeling has not yet been elucidated. The objective is to evaluate the relationship between versicanases, inflammatory tumor microenvironment and the development of metastases in a 4T1 murine model. Histomorphological analyzes were performed on primary tumor, regional lymph node and lung samples in the groups of 7, 14, 21 and 28 days of tumor growth and analyzes of the immunohistochemical expression of VCAN, VKINA, disintegrins and metalloproteinases with thrombospondin domains (ADAMTS) - 1, -5 and -15, as well as analysis of the immunophenotypic profile of immune response cells in peripheral blood and tumor infiltrate. Over the 28 days, there was an increasing increase in VCAN expression. However, VKINA expression reached a higher median at 21 days of tumor growth, as did ADAMTS15. ADAMTS1 and 5 were more expressed in stroma at 21 and 28 days of tumor inoculation. VCAN, ADAMTS 1 and 5 showed greater expression in the last weeks of the experiment, with a direct correlation to the predominantly pro-inflammatory profile. However, VKINA and ADAMTS15 showed greater expression in the third week, when metastases appear in this model In addition to the high expression of VKINA in 21 days, this proteolyte was related to a predominantly anti-inflammatory immune profile, creating a microenvironment favorable to tumor progression. The results suggest a relationship between the degradation of the extracellular matrix (ECM) and the inflammatory profile. However, it is still necessary to understand the effects of these immune profiles on the remodeling of the extracellular matrix, particularly concerning versicanases and their proteolysis at different stages of tumor progression
