Abstract in Portuguese | protozoários tripanosomatídeos incluem patógenos de importância médico veterinária entre os quais se encontra o Trypanosoma brucei, espécie alvo deste estudo. A regulação gênica nesses organismos é mediada por mecanismos pós transcricionais, tais como o controle da estabilidade e tradução dos mRNAs. Em eucariotos, a regulação da tradução ocorre principalmente na etapa de iniciação, mediada por fatores de iniciação eucarióticos (eIFs). O complexo eIF4F, composto pelas subunidades eIF4A, eIF4E e eIF4G, é o responsável pelo reconhecimento dos mRNAs, com a subunidade eIF4E se ligando a extremidade 5’ do mRNA. O eIF4E possui seis homólogos já identificados nos tripanosomatídeos (EIF4E1 ao E6). O EIF4E2 é o menos conhecido deles, porém estudos mais recentes demostraram que este é capaz de se ligar de forma específica a um de dois homólogos da proteína SLBP (stem loop binding protein) de tripanosomatídeos, a SLBP2. Na maior parte dos eucariotos um único homólogo de SLBP é encontrado e em metazoários demostrou-se ter um papel na regulação do metabolismo de mRNAs de histonas. Esse estudo visa avançar no estudo do EIF4E2 e SLBPs através de duas abordagens principais: quantificação dos seus níveis intracelulares em células procíclicas de T. brucei; avaliação de proteínas parceiras associadas, através de reações de imunoprecipitação seguida de identificação por espectrometria de massas. A quantificação da expressão de ambas as proteínas foi realizada, confirmando um perfil de expressão da SLBP1, expresso constitutivamente, significativamente maior que a SLBP2, diferencialmente expressa durante o ciclo celular. Resultados de espectrometria demonstraram a associação do complexo de MKT-1 co-precipitando de forma específica com a SLBP1, sugerindo um possível papel funcional na ativação da tradução nesses organismos, enquanto que a SLBP2 e EIF4E2 aparentemente não estão associados diretamente a tradução, com a SLBP2 aparentemente se associando a um sistema de regulação envolvendo proteínas do sistema ubiquitina-proteassoma, influenciando na regulação do seu metabolismo proteico. Os resultados obtidos podem embasar futuros estudos que busquem métodos mais efetivos de combate para patologias causadas pelo T. brucei e organismos relacionados, como o T. cruzi e espécies de Leishmania. | en_US |

Abstract | Trypanosomatid protozoa include pathogens of medical-veterinary importance, among which is Trypanosoma brucei, the target species of this study. Gene regulation in these organisms is mediated by post-transcriptional mechanisms, such as control of the stability and translation of mRNAs. In eukaryotes, translation regulation occurs mainly in the initiation step, mediated by eukaryotic initiation factors (eIFs). The eIF4F complex, formed by the eIF4A, eIF4E and eIF4G subunits, is responsible for the recognition of mRNAs, with the eIF4E subunit binding to the 5' end of the mRNA. eIF4E has six homologues already identified in trypanosomatids (EIF4E1 to E6). EIF4E2 is the least known of these, but more recent studies have shown that it is able to specifically bind to one of two homologues of the trypanosomatid SLBP (stem loop binding protein), SLBP2. In most eukaryotes a single homolog of SLBP is found and in metazoans a function in the regulation of histone mRNA metabolism has been shown. This study aimed to advance the study of EIF4E2 and SLBPs through two main approaches: quantification of their intracellular levels in T. brucei procyclic cells; evaluation of associated partner proteins, through immunoprecipitation reactions followed by identification through mass spectrometry. The quantification of the expression of both proteins was realized, confirming an expression profile of SLBP1, constitutively expressed, significantly higher than SLBP2, expressed differentially during the cell cycle. Spectrometry results demonstrated the association of the MKT-1 complex coprecipitating specifically with SLBP1, suggesting a possible functional role in the activation of translation in these organisms, while SLBP2 and EIF4E2 are apparently not directly associated with translation, with SLBP2 apparently associating with a regulatory system involving proteins of the ubiquitin-proteasome system, influencing the regulation of its protein metabolism. The obtained results can support future studies that seek more effective methods of controlling pathologies caused by T. brucei and related organisms, such as T. cruzi and Leishmania species. | en_US |
