O vírus Zika (ZIKV) demonstrou, nos últimos anos, um relevante potencial de iniciar epidemias. Devido à grande quantidade de casos de ZIKV confirmados em vários países em todo mundo e a sua relação com distúrbios neurológicos, ficou evidente a importância de se estudar os mecanismos envolvidos na infecção viral. Entretanto, as vias de sinalização envolvidas na resposta imune contra o ZIKV ainda não estão bem elucidadas. O presente estudo avaliou a ativação das vias de sinalização JAK/STAT e MAPK em células monocíticas (THP-1) infectadas com ZIKV. Foi demonstrado que células THP-1 são suscetíveis a infecção do ZIKV e apresentam um perfil de morte celular intensa após 48 horas de infecção. Uma vez infectadas pelo ZIKV, a expressão gênica de PI3K, Akt e NF-κB é aumentada, a fosforilação de p38MAPK, ERK1/2, STAT1, STAT3, STAT5 e STAT¨6 é induzida e a produção de IL-6, IL-8, CXCL9, CCL2, CCL5 e CXCL10 é ativada, havendo uma modulação imunológica de acordo com o tempo pós infecção. Além disso, a inibição de p38MAPK induziu a diminuição dos níveis de IL-8 e CXCL10, sugerindo que a regulação dessas citocinas dependem, parcialmente, da via de sinalização MAPK. Mais estudos são necessários para caracterizar as vias de sinalização da imunidade inata frente a infecção do ZIKV .
Abstract
Zika virus (ZIKV) has shown a relevant potential for initiating epidemics in recent years. Due to the large number of confirmed ZIKV cases in various countries around the world and their relationship to neurological disorders, the importance of studying the mechanisms involved in viral infection has become evident. However, the signaling pathways involved in the immune response against ZIKV are not yet well understood. The present study evaluated the activation of JAK / STAT and MAPK signaling pathways in ZIKV-infected monocytic cells (THP-1). THP-1 cells have been shown to be susceptible to ZIKV infection and to have an intense cell death profile 48 hours after infection. Once infected with ZIKV, PI3K, Akt and NF-κB gene expression increases, phosphorylation of p38MAPK, ERK1 / 2, STAT1, STAT3, STAT5 and STAT¨6 is induced, andIL-6 production is activated. IL-8, CXCL9, CCL2, CCL5 and CXCL10 with immunological modulation according to time after infection. In addition, inhibition of p38MAPK induced a decrease in IL-8 and CXCL10 levels, suggesting that the regulation of these cytokines partly depends on the MAPK signaling pathway. Further studies are needed to characterize innate immunity signaling pathways against ZIKV .
