ESTABELECIMENTO DE CULTIVO PRIMÁRIO DE RHODNIUS PROLIXUS (HEMIPTERA: TRIATOMINAE)

Triatomíneos são insetos hematófagos e vetores de tripanossomas, sendo a linhagem celular uma ferramenta para elucidação da interação entre células e a propagação desses patógenos. Objetivou-se nesse trabalho estabelecer o cultivo primário de células proveniente de ovos, da glândula salivar e do intestino de Rhodnius prolixus. Este estudo foi conduzido no Laboratório Interdisciplinar de Vigilância Entomológica em Diptera e Hemiptera (LIVEDIH), da Fundação Oswaldo Cruz, em parceria com o Laboratório de Doenças Parasitárias (LDP), da Universidade Federal Rural do Rio de Janeiro. Ninfas de quinto instar de R. prolixus foram obtidas da criação de triatomíneos do LIVEDIH. Após atingirem a fase adulta foram alimentados por uma hora e separados por casal. Os ovos provenientes da cópula foram retirados recém ovipostos ou com até sete dias. Após a coleta dos ovos, os insetos foram dissecados para obtenção dos órgãos (glândula salivar e intestino). Cada material biológico foi lavado, separadamente, e, após, macerado em meio Leibovitz´s L15-B suplementado. O conteúdo de cada macerado foi filtrado e transferido para quatro frascos de cultivo de 25 cm2 , sendo dois frascos mantidos em estufas com temperatura de 28ºC e dois à 32ºC. Estas foram acompanhadas diariamente nos sete primeiros dias e após esse período acompanhadas semanalmente A partir do segundo dia do início da realização do cultivo com os ovos de R. prolixus foi observada a fixação da maioria das células na superfície do frasco com inúmeros agregados celulares, sendo gradativamente observado o aumento da confluência da monocamada indicando o crescimento celular, quando submetidas a 28ºC. Após o sétimo dia da fixação foi observado uma grande variedade de tipos celulares que se diferenciaram em células fibroblastoides e, posteriormente, em células epitelioides arredondadas. Não houve aderência celular nos frascos submetidos à temperatura de 32ºC. As células da glândula salivar aderiram ao frasco de cultivo quando submetidas à temperatura de 28ºC, mas não foi observado crescimento celular, sendo este considerado cultivo primário temporário. Quando submetidas à temperatura de 32ºC, não foi observado aderência celular. Em relação ao cultivo primário proveniente de células do intestino foi observada contaminação bacteriana nos frascos submetidos em ambas às temperaturas, após 24 horas. O cultivo de células dos ovos de R. prolixus permanecem viáveis em cultura há 583 dias, sob a temperatura de 28ºC, e encontra-se na sexta passagem celular. Neste trabalho foi possível, pela primeira vez no Brasil, o estabelecimento de cultura primária de ovos de R. prolixus, o que pode auxiliar em futuros estudos de interação vetor-patógeno. Contudo, busca-se o estabelecimento de uma linhagem proveniente desse cultivo
Abstract
Triatomines are hematophagous insects and trypanosome vectors, and the cell line is a tool for elucidating the interaction between cells and the propagation of these pathogens. The objective of this work was to establish the primary cells culture from eggs, the salivary gland and the intestine of Rhodnius prolixus. This study was conducted at the Interdisciplinary Laboratory for Entomological Surveillance in Diptera and Hemiptera (LIVEDIH), of the Oswaldo Cruz Foundation, in partnership with the Laboratory of Parasitic Diseases (LDP), of the Federal Rural University of Rio de Janeiro. Fifth instar nymphs of R. prolixus were obtained from the breeding of LIVEDIH triatomines. After reaching adulthood, they were fed for an hour and separated by couple. The eggs from the copulation were removed freshly laid or up to seven days old. After collecting the eggs, the insects were dissected to obtain the organs (salivary gland and intestine). Each biological material was washed separately and, afterwards, macerated in supplemented Leibovitz´s L15-B medium. The contents of each macerate were filtered and transferred to four 25 cm2 culture flasks, two flasks kept in greenhouses with a temperature of 28ºC and two at 32ºC. These were followed up daily for the first seven days and after that period, followed up weekly From the second day of the beginning of the culture with the eggs of R. prolixus, the fixation of the majority of the cells on the surface of the flask with numerous cell aggregates was observed, being gradually observed the increase of the confluence of the monolayer indicating the cell growth, when subjected to 28ºC. After the seventh day of fixation, a wide variety of cell types were observed, which differentiated into fibroblastoid cells and, subsequently, into rounded epithelioid cells. There was no cell adhesion in the flasks submitted to a temperature of 32ºC. The cells of the salivary gland adhered to the culture flask when subjected to a temperature of 28ºC, but no cell growth was observed, which was considered a temporary primary culture. When subjected to a temperature of 32ºC, cell adhesion was not observed. Regarding the primary culture from intestinal cells, bacterial contamination was observed in the flasks submitted to both temperatures, after 24 hours. The cultivation of cells of R. prolixus eggs remained viable in culture for 583 days, under the temperature of 28ºC, and is in the sixth cell passage. In this work, it was possible, for the first time in Brazil, to establish a primary culture of R. prolixus eggs, which may help in future studies of vector-pathogen interaction. However, it is sought to establish the cell line from this cultivation
