INTRODUÇÃO As leishmanioses compreendem um conjunto de doenças infecciosas causadas por parasitos do gênero Leishmania que atingem milhares de pessoas no mundo todos os anos. O tratamento convencional ainda é altamente tóxico, prolongado, financeiramente dispendioso e está associado à falha terapêutica, justificando a pesquisa e o desenvolvimento de novos medicamentos. OBJETIVO. No presente estudo foi avaliada a atividade anti-Leishmania de 46 compostos químicos a partir da aplicação de um método de análise colorimétrico. MÉTODOS. A padronização do ensaio colorimétrico semi-automatizado para medir a viabilidade dos parasitos foi realizada utilizando macrófagos J774 e a cepa BA-125 de L. amazonensis, tratando ou não com anfotericina B, seguida de incubação a 25 oC em meio Schneider e a quantificação dos promastigotas com AlamarBlue® e leitura da absorbância a 570 e 600 nm. Em seguida, avaliamos a citotoxicidade de 46 compostos (26 hidrazonas, o lapachol livre, três complexos contendo rutênio e o precursor do complexo, a mefloquina e 5 complexos contendo 4-aminoquinolina, a lausona livre e um composto com rutênio, a alizarina livre e um composto com rutênio, um complexo contendo atovaquona, três compostos tiossemicarbazonas e o ligante livre), em células de mamífero não infectadas. O potencial leishmanicida dos compostos em estudo foi avaliado no ensaio colorimétrico padronizado. RESULTADOS. Não houve diferença estatística entre os valores de IC50 encontrados pelo método da contagem manual de promastigotas e o ensaio de AlamarBlue® utilizando anfotericina (p = 0,3429). Dois compostos dos 26 testados da classe das hidrazonas (MM20 e MM24) apresentaram a atividade anti-Leishmania superior a 70% de inibição, com valores de CC50 112,3 e 120 \03BCM, IC50 23,76 e 19,56 \03BCM e IS 6,13 e 4,72, respectivamente. As tiossemicarbazonas foram os compostos menos tóxicos, porém com nenhum composto com atividade anti-Leishmania na maior concentração testada. Os complexos contendo 4-aminoquinolinas foram os mais citotóxicos (CC50 de 12,7 a 18,2 \03BCM), e três compostos (WV-65, WV-86, WV-74) tiveram maior atividade anti-Leishmania (IC50 de 3,2 a 12 \03BCM e IS de 1,72 a 8,34). CONCLUSÃO. O ensaio semiautomatizado pode ser útil para a triagem de compostos anti-Leishmania, e identificou quatro compostos mais promissores (WV-65, WV-86, WV-74 e MM20), pertencentes às 4-aminoquinolinas e hidrazonas.
Abstract
INTRODUCTION. Leishmaniasis comprises a group of infectious diseases caused by parasites of the Leishmania genus that affect thousands of people worldwide every year. Conventional treatment is still highly toxic, prolonged, financially expensive and is associated with therapeutic failure. Therefore, research and development of new drugs to treat leishmaniasis is justified. OBJECTIVE. In the present study, the anti-Leishmania activity of 46 chemical compounds was evaluated using a colorimetric assay method. METHODS. The standardization of the semi-automated colorimetric test to measure the viability of the parasites was performed using J774 macrophages and the BA-125 strain from L. amazonensis, treating or not with amphotericin B, followed by incubation at 25 °C in Schneider medium and the quantification of promastigotes with AlamarBlue® and absorbance reading at 570 and 600 nm. Then, we evaluated the cytotoxicity of 46 compounds (26 hydrazones, free lapachol, three complexes containing ruthenium and the complex precursor, mefloquine and 5 complexes containing 4-aminoquinoline, free lausone and a compound with ruthenium, free alizarin and a compound with ruthenium, a complex containing atovaquone, three thiosemicarbazone compounds and the free ligand) in uninfected mammalian cells. The leishmanicidal potential of the compounds under study was evaluated in the standardized colorimetric assay. RESULTS. There was no statistical difference between the IC50 values found by the method of manual counting of promastigotes and the AlamarBlue® assay using amphotericin (p = 0,3429). Two compounds of the 26 hydrazones (MM20 and MM24) showed anti-Leishmania activity greater than 70% of inhibition, with CC50 values of 112.3 and 120 \03BCM, IC50 23.76 and 19.56 \03BCM and IS 6.13 and 4.72, respectively. Thiosemicarbazones were the least toxic compounds, but with no compound with anti-Leishmania activity in the highest concentration tested. The complexes containing 4- aminoquinolines were the most cytotoxic (CC50 from 12.7 to 18.2 \03BCM), and three compounds (WV-65, WV-86 and WV-74) had the highest anti-Leishmania activity (IC50 of 3.2 to 12 \03BCM and IS from 1.72 to 8.34). CONCLUSION. The semiautomated assay can be useful for the screening of anti-Leishmania compounds, and identified four more promising compounds (WV-65, WV-86, WV-74 and MM20), belonging to 4-aminoquinolines and hydrazones.
