Abstract in Portuguese | O neuropeptídeo Y (NPY) é um dos neurotransmissores mais abundantes do sistema nervoso, que pode estimular angiogênese, inflamação e adipogênese na região abdominal através do Receptor 2 do NPY (NPYR2). Alterações na sua sinalização foram associadas, com a Síndrome Coronariana Aguda (SCA). O estudo avaliou a relação entre as variantes. c.20T>C, c.84G>A, c.150G>A, c.-485T>C do NPY e as c.-1088C>T e c.-1116A>G do NPYR2, como da metilação de DNA global, com a gravidade da SCA, através do Thrombolysis in Myocardial Infarction (TIMI) score. Participaram do estudo 221 pacientes com SCA e 95 sem SCA, selecionados no Real Hospital Português , situado em Recife, PE. Além, de 278 doadores de sangue da Fundação de Hematologia e Hemoterapia, Recife, PE. Houve associação do gênero masculino (p = 0,0014; OR = 2,42) tabagismo (p = 0,0291; OR = 2,31) e dislipidemia (p < 0,0001; OR = 3,76) com a SCA. Existiu associação da variante c.20T>C do NPY com SCA (p = 0,0274; OR = 3,24), entre o grupo com SCA e o grupo saudável. Na análise entre as variantes e os fatores de risco no grupo com SCA, NPY c.84G>A foi associado com a hipertensão. A análise entre o TIMI e as variantes mostrou para a NPY c.20T>C (p = 0,0261; OR = 4,00) uma diferença entre os grupos TIMI baixo e intermediário/alto. Ocorreu um maior nível de metilação no grupo SCA e quando estratificado pelo TIMI, o grupo intermediário/alto apresentou um menor padrão de metilação. A variante NPY c.20T>C ao mesmo tempo em que pode ser um fator de risco para SCA pode influenciar com um efeito de proteção na gravidade da . Um perfil hipermetilado de DNA global no grupo com SCA foi associado ao risco de SCA, podendo se tornar um biomarcador para SCA. Em relação ao escore de risco TIMI, o baixo nível de metilação global se mostrou associado ao grupo intermediário/alto para SCA, possibilitando esse perfil hipometilado ser utilizado como um marcador para estratificar os pacientes para risco de morte ou de eventos cardiovasculares. | pt_BR |

Abstract | The neuropeptide Y (NPY) is one of the most abundant neurotransmitters in the nervous system, which can stimulatte angiogenesis, inflammation and adipogenesis in the abdominal region through NPY Receptor 2 (NPYR2). Alterations in its signaling were associated with Acute Coronary Syndrome (ACS). The study evaluated the relationship between variants c.20T>Cc.84G>A), c.150G>A , c.-485T>C of NPY and c.-1088C>T and c.-1116A>G of NPY2R, as well as of global DNA methylation, with the severity of SCA, through the Thrombolysis in Myocardial Infarction (TIMI) score. A total of 221 patients with ACS and 95 patients without ACS, all participated in the study from the Real Hospital Português, located in Recife, PE. In addition, of 278 blood donos of Fundação de Hematologia e Hemoterapia, Recife, PE. . There was an association among male gender ((p = 0.0001; OR = 2.42), smoking ((p = 0.0029; OR = 2.31) and dyslipidemia (p < 0.0001; OR = 3.76) with ACS. Also, an association of variant c.20T>C (p = 0,0274; OR = 3,22), of NPY with SCA, between the group with ACS and the healthy group. In the analysis between variants and risk factors in the ACS group, NPY c.84G>A was associated with hypertension. The analysis between TIMI risk and the variants showed for NPY c.20T>C a significant difference between the low and intermediate/high TIMI risk groups (p = 0.0223; OR = 4.00). A higher methylation level was observed in the SCA group. When stratified by TIMI risk, the intermediate/high group presented a lower methylation pattern. The variant NPY c. 20T>C while it may be a risk factor for SCA it may influence with a protective effect to the severity of the disease. A hypermethylated global DNA profile in group with ACS may be associated with the risk of ACS, and may become a promising biomarker for ACS. Regarding the TIMI risk score, the low level of global methylation was most associated with the intermediate / high risk group for ACS, allowing this hypomethylated profile to be used as a marker to stratify patients to risk of death or cardiovascular events. | pt_BR |
