IMUNOPATOLOGIA DE ISOLADOS DE LEISHMANIA (VIANNIA) BRAZILIENSIS EM GOLDEN HAMSTER (MESOCRICETUS AURATUS)

A leishmaniose tegumentar americana (LTA) causada por Leishmania (Viannia) braziliensis é uma doença inflamatória crônica amplamente distribuída no Brasil, podendo apresentar diversas formas clínicas. Em um foco endêmico no estado de Minas Gerais, a diversidade genética deste parasito foi associada a manifestações clínicas atípicas nos pacientes portadores da LTA. Baseado neste achado, propusemos estudar o comportamento imunopatológico de cinco isolados de L. (V.) braziliensis em
hamster dourado. Infecções experimentais foram realizadas utilizando 1x106 parasitos por inóculo por via intradérmica. Foram avaliados parâmetros relacionados às lesões cutâneas, como tempo de surgimento, aspecto, tamanho, histopatologia, carga parasitária e expressão de mRNA de diferentes citocinas e quimiocinas nas lesões cutâneas, trinta e sessenta dias pós-infecção (PI). Os resultados revelaram a ocorrência de padrões distintos em relação ao tempo de surgimento, tamanho e carga parasitária
das lesões. Diferenças histopatológicas foram observadas com relação à extensão e densidade do infiltrado inflamatório, apesar da composição celular semelhante entre as cepas. Padrões distintos de resposta imune foram encontrados principalmente na fase inicial da infecção (trinta dias PI). Neste período, as variantes genéticas induziram a expressão altos níveis de tnf e il12 enquanto o isolado não variante induziu a expressão de ifn , il6, il4 e il10. Sessenta dias PI, diferentes padrões na expressão de citocinas e
quimiocinas foram observados em hamsters infectados com as cepas variantes, enquanto reduções significativas nas expressões de ifn , il6, ccl17 e ccl22 foram observadas nos animais infectados com o isolado não variante. Utilizando uma análise multivariada, foi possível segregar os isolados em dois clusters, no qual os isolados variantes genéticos de L. (V.) braziliensis se agruparam de forma independente do isolado não variante, sugerindo que os padrões distintos no comportamento imunobiológico podem estar associados à diversidade genética previamente descrita
para estes isolados, que por sua vez, poderiam ser capazes de modular vias distintas de resposta imune.
Abstract
American cutaneous leishmaniasis (ACL) caused by Leishmania (Viannia) braziliensis is a chronic inflammatory disease widely spread in Brazil, which may present several clinical manifestations. In an endemic focus in the State of Minas Gerais, the genetic diversity of this parasite has been associated to atypical clinical manifestations in ACLpatients. Based on this report, we propose to evaluate the immunobiological behavior of genetic variant strains of L. (V.) braziliensis in golden hamster. Experimental infections were standardized with 1x106 parasites per inocula. Characteristics of skin lesions, histopathological features and parasite burden were independently-analyzed at thirty and sixty days post-infection (PI). In addition, detailed analyses of the mRNA expression of different cytokines and chemokines in hamster-skin lesions were performed. The results revealed the occurrence of distinct patterns in the onset time, lesion size and parasite burden among the strains. Differences in the extension and composition of the inflammatory infiltrate were also observed, although the cellular composition of the granulomas appeared similar in all infected hamsters. Distinct patterns of immune response were found mainly in the early-phase disease (thirty days
PI). At this time point, the genetic variant strains expressed high levels of tnf and il12, whilst the non-variant strain expressed high levels of ifn , il6, il4 and il10. At sixty days PI, distinct patterns of cytokines and chemokines expressions were observed for the variant strains, whilst significant reductions in the expression of ifn , il6, ccl17 and ccl22 were observed for the non-variant strain. The multivariate analysis segregated the strains in two clusters, in which the genetic variant strains of L. (V.) braziliensis did not
group with the non-variant strain, suggesting that the distinct patterns in the
immunobiological behavior may be related with the known genetic diversity previously described to these strains, which are able to trigger distinct pathways of immune response.
