AVALIAÇÃO DO POTENCIAL ADJUVANTE DE LECTINAS VEGETAIS NA VACINAÇÃO CONTRA LEISHMANIOSE CUTÂNEA EXPERIMENTAL

As leishmanioses são causadas por parasitas do gênero Leishmania, importantes parasitas intracelulares de macrófagos que se manifestam em formas clínicas diferentes. A resposta imune celular é a resposta protetora efetiva contra parasitas intracelulares. Já foi demonstrado que algumas lectinas de plantas foram capazes de induzir a produção, por células mononucleares murinas, de óxido nítrico (NO), interferon gama (IFN-y) e IL-12, moléculas importantes em uma resposta celular. No presente trabalho nós investigamos o potencial imunoestimulatório de três lectinas vegetais e avaliamos a possibilidade de utilizá-las como adjuvantes em um modelo de vacinação contra leishmnaniose cutânea experimental. Para avaliar o potencial estimulatório das lectinas, células esplênicas murinas e CMSP humanas foram estimuladas em cultura com as lectinas Canavalia brasiiiensis (ConBr), Pisum arvense (PAA) ou Artocarpus integrifolia (KM(+)) e os sobrenadantes coletados para determinação da produção de IFN-y, IL-10 e IL-4. Todas as lectinas foram capazes de induzir a secreção de IFN-y e uma pequena quantidade de IL-4 por células esplênicas murinas e IFN-y, IL-10 e IL-4 por CMSP humanas. Para avaliação do potencial adjuvante, camundongos C57BL/6 e BALB/c foram imunizados com as lectinas ConBr, PAA ou KM(+) com o antigeno solúvel de Leishmania amazonensis (SLA) ou com o antigeno ou lectinas isoladamente. Antes do desafio com Leishmania amazonensis BA125, os soros dos animais imunizados foram coletados para determinação de IgG ant\-Leishmania. A avaliação da infecção foi monitorada semanalmente pela medida da pata infectada. Apenas os animais imunizados com a lectina PAA e PAA+SLA produziram altos títulos de IgG anW-Leishmania. A resposta celular foi avaliada por uma reação de hipersensibilidade tardia (DTH), realizada dez semanas após o desafio. Não foi possível relacionar a resposta de DTH a um perfil de resistência. Apenas os animais imunizados com a lectina KM(+) foram protegidos parcialmente contra posterior infecção. Apesar das lectinas ConBr, PAA and KM(+) terem demonstrado estimular a secreção de moléculas capazes de direcionar uma resposta imune celular, nossos resultados sugerem que as lectinas testadas não possuem atividade adjuvante na vacinação contra Leishmania amazonensis.
Abstract
Leishmaniasis is caused by parasites of the genus Leishmania, intracellular parasites of macrophages that cause a great variety of diseases. A cell-mediated immune response has shown to be protective against intracellular parasites. It has been demonstrated that plant lectins are capable of inducing nitric oxide (NO), gamma interferon (IFN-y) and interleukin 12 (IL-12) production, which are key molecules in a cellular immune response by murine mononuclear cells. In the present work, we investigate the possibility of using lectins from Canavalia brasiiiensis (ConBr), Pisum arvense (PAA) or Artocarpus integrifolia (KM(+)) as adjuvants to protect against Leishmania infection. Murine spleen cells and human PBMC were stimulated in culture with one of the lectins and their supernatants were collected for IFN-y, IL-10 and IL-4 determination. All lectins were capable of inducing IFN-y, but a low IL-4 secretion from murine spleen cells. The lectins tested were also capable of inducing IFN-y, IL-10 and IL-4 secretion from human PBMC. For evaluation of adjuvant potential C57BL/6 and BALB/c mice were immunized with ConBr, PAA or KM(+) lectins, in the presence or absence of soluble Leishmania amazonensis antigen (SLA). Before challenge with Leishmania amazonensis BA125 strain, serum was obtained for ant\-Leishmania IgG level determination. The evolution of infection was monitored by weekly measurements of footpad thickness. Only the animals immunized with PAA lectin and PAA+SLA produced higher titers of specific IgG. Moreover, no protection of the mice immunized with either ConBr or PAA lectins was achieved. Delayed type hipersensitivity (DTH) was also assessed ten weeks after challenge. The DTH response could not be related to a resistance profile in the immunized animals. Only the animals immunized with KM(+) lectin alone were partialy protected against further infection. Although, ConBr, PAA and KM(+) lectins have shown to induce the production of molecules capable of directing a cell- mediated immune response, our results suggest that the lectins tested are not useful adjuvants to vaccination against Leishmania amazonensis.
