AVALIAÇÃO DO PAPEL DO INTERFERON-Y NA MODULAÇÃO DA RESPOSTA TECIDUAL DURANTE A INFECÇÃO PELO MYCOBACTERIUM BOVIS

O IFN-y tem sido implicado na defesa do hospedeiro contra as micobactérias. Esta citocina, produzida e liberada por linfócitos T recrutados para o sítio da infecção, é considerada o agente chave da ativação endógena, promovendo efeitos antimicobacterianos de macrófagos. Diversos modelos experimentais têm sido desenvolvidos para delinear o papel de citocinas responsáveis pelo controle da tuberculose. O presente estudo visa delinear o papel do IFN-y na modulação da resposta inflamatória granulomatosa no tecido hepático e pulmonar em camundongos infectados experimentalmente com o M. bovis. Os experimentos foram realizados utilizando-se camundongos com deficiência do gene responsável pela produção do IFN-y (C57BL/6 IFN-y-l-) e camundongos que produzem IFN-y (C57BL/6 IFN-y+l+), ambos infectados por via intravenosa com M. bovis. Avaliou-se a capacidade de sobrevida e multiplicação do M. bovis no fígado e pulmões, assim como características histopatógicas nestes dois órgãos. No curso da infecção (15, 30, 50 e 100 dias pós-infecção) a CFU (unidade formadora de colônia) foi quantificada no tecido hepático e pulmonar dos dois grupos de camundongos. A carga bacilar dos animais IFN-y-l- foi significantemente maior que nos IFN-y+/+. A quantidade de CFU foi maior no fígado que nos pulmões dos animais IFN-y-l- durante todo o estudo, enquanto nos IFN-y+/+ apenas no primeiro ponto estudado (15 dias pós-infecção). Adicionalmente, nos camundongos IFN-y/- ocorreu um aumento contínuo da carga bacilar com o transcurso da infecção, enquanto os camundongos IFN-y+/+ foram capazes de reduzir a carga bacilar no tecido hepático nos períodos mais tardios da análise. As alteraíões histopatológicas começam mais tardiamente nos camundongos IFN-y-/- (30 dias de pós-infecção). Estes formam granulomas sempre em menor número e tamanho que os IFN-y+/+. As alterações granulomatosas no fígado persistem até os 100 dias no camundongo IFN-y-l- e diminuem nos IFN-y+/+. Portanto, a presença do IFN-y influenciou no sobrevida e multiplicação do M. bovis no fígado e pulmões dos camundongos, assim como na formação e resolução do processo granulomatoso.
Abstract
IFN-y has been largely implicated in tlie liost defense against mycobacteria. This cytokine, produced and released by T lymphocytes recruited to the site of infection, is considered the key agent of endogenous activation, promoting antimicrobial effects on macrophages. Diverse experimental models have been developed to delineate the role of cytokines responsible for the control of tuberculosis. The present study aims to evaluate the role of IFN-y in the immune granulotamous response against M. bovis in liver and lung tissue. The experiment utilized C57BL/6 mice iacking the genes for synthesis of IFN-y (IFN-y-/-) and their wild-type counterparts (IFN-y+/+), that were intravenously infected with M. bovis. The ability of /W. bovis to survive and reproduce in the liver and lungs was evaluated, as well as characterized histopathologically in these two organs. In the course of infection (15, 30, 50, and 100 days post-infection), baciilary load (number of colony forming units) in the liver and lungs was quantified in the two groups of mice. Baciilary load was greater in both organs in the animals that were IFN-y-/- than in mice that were IFN-y+/+. Baciilary load was greater in the liver than in the lungs of IFN-y-/- mice, during the entire study period, while in controls this was true only at the first time-point (15 days post-infection). Baciilary load increased progressively in the two organs of the IFN-y'-/- mice, whereas wildtype animals were of controling baciilary growth. The granulomas were observed in the IFN-y-/- mice after 30 days of infection, and earlier in controls. Granulomatous changes in the liver persisted in the IFN-y-/- mice and resolved in the controls, more significantly after 100 days. Additionally, IFN-y -/- mice had fewer and smaller granulomas than IFN-y+/+. The presence of IFN-y influenced the multiplication of M. bovis in the liver and lungs of mice, as well as the formation and resolution of granulomatous process.
