Lutzomyia migonei (França 1920) é um vetor conhecido de Leishmania braziliensis, agente causador da leishmaniose cutânea na América do Sul. Recentemente, foi sugerido que esta espécie também pode transmitir Leishmania infantum, contribuindo nas infecções por leishmaniose visceral. Neste estudo avaliamos pela primeira vez os aspectos biológicos, morfológicos e genéticos de diferentes populações de Lu. migonei no Brasil. Não foram observadas diferenças morfológicas ou morfométricas significativas entre as populações estudadas. No entanto, ao analisar a variabilidade genética verifiou-se a existência de dois clados monofiléticos irmãos. Ao analisar a ultraestrutura de Lu. migonei verificou-se a presença de estruturas disseminadoras de feromônio distribuídas em diferentes segmentos abdominais dos machos e que a produção de feromônio aumenta gradualmente com a maturação das células glandulares, os quais parecem estabilizar em insetos adultos mais maduros em torno de três a sete dias pós-emergência, coincidindo com o período em que os machos são mais capazes de atrair as fêmeas. Entender o comportamento biológico e observar as evidências para comunicação de feromônio poderá ajudar a compreender o papel dos feromônios sexuais neste importante vetor. Ademais, o uso de marcadores moleculares (p.ex.: citocromo oxidase subunidade I) poderá fornecer informações valiosas sobre a genética de diferentes populações de Lu. migonei e sobre uma possível relação entre a variabilidade genética e a competência vetorial e/ou produção de diferentes tipos de feromônios
Abstract
The sand fly Lutzomyia migonei (França 1920) is a known vector of Leishmania braziliensis, the causative agent of cutaneous leishmaniasis in South America. Recently it has been suggested that this specie can also transmit Le. infantum thus contributing to the burden of visceral leishmaniasis infection. This study describes for the first time the biological,
morphological and genetic aspects of different Lutzomyia migonei populations of Brazil. No significant morphological or morphometric differences between all populations were observed. However, when analyzing the genetic variability we found the stocks of two close
monophyletic clades. Analyzing the ultrastructure of Lu. migonei verified the presence of disseminating pheromone structures distributed in different abdominal segments of the male and the pheromone production gradually increases with maturation of glandular cells, which appeared to stabilize in mature adult insects around three to seven days post emergence,
coinciding with the period in which males are able to attract females. Understanding the biological behavior and observe the evidence for pheromone communication can help to understand the role of sex pheromones in this important vector. Furthermore, the use of
molecular markers (i.e.: cytochrome oxidase subunit I) can provide valuable information about the genetics of different populations of Lu. migonei and the possible relationship between genetic variability and vector competence and / or production of different types of pheromones.
